硝苯地平对大鼠肺动脉平滑肌细胞增殖的影响

摘    要:目的研究钙拮抗剂硝苯地平对肺动脉平滑肌细胞增殖的影响 ;探讨钙离子拮抗剂对肺血管构型重建的抑制作用。方法分离培养大鼠肺动脉平滑肌细胞 ;用MTT法检测细胞增殖 ;用流式细胞仪分析细胞周期。结果硝苯地平能剂量依赖地抑制血清诱导的肺动脉平滑肌细胞增殖 ,对照组的增殖指数和S期细胞分数为 36 0 2 %和 15 88% ;硝苯地平 1× 10 -8mol·L-1、1× 10 -7mol·L-1、1× 10 -6mol·L-1组的PI和SPF分别为 31 16 %、2 6 94 %、2 4 95 %和 13 10 %、11 4 7%、9 6 0 %。结论肺动脉平滑肌细胞增殖中有钙离子信号的参与 ;硝苯地平能够抑制血清诱导的肺动脉平滑肌细胞增殖 ;硝苯地平对肺血管构型重建可能有一定的抑制作用

Abstract:Objective To study the effects of nifedipine,a calcium antagonist on the proliferation of pulmonary artery smooth muscle cells and investigate the effect of nifedipine on pulmonary remodeling.Methods The pulmonary artery smooth muscle cells were isolated from the rat pulmonary artery.Cultured pulmonary artery smooth muscle cells were evaluated by the MTT assay.Proliferation index (PI) and S-phase cell fraction (SPF) were performed by the flow cytometry.Results Nifedipine dose-dependently inhibited pulmonary artery smooth muscle cells proliferation was induced by fetal bovine serum.The proliferation index of cells treated with nifedipine 1×10~(-8),1×10~(-7),1×(10~(-6) mol·L~(-1)) were 31.16%,26.94%,24.95%; s-phase cell fractions were 13.10%,11.47%,9.60% respectively.PI and SPF in the control was 36.02% and 15.88%.Nifedipine inhibited the cell cycle from the G_0/G_1 phase into S-phase.Conclusions Nifedipine blocked pulmonary artery smooth muscle cells mitogenesis induced by serum.Ca~(2+) and Ca~(2+) channel played important roles in the proliferation of pulmonary artery smooth muscle cells and pulmonary remodeling.Nifedipine may have inhibitory effect on pulmonary remodeling.
