白藜芦醇对大鼠脊髓损伤早期凋亡的影响

摘    要:目的:探讨脊髓损伤后使用白藜芦醇(RES)对脊髓损伤早期神经细胞凋亡的影响。方法:将62只Spra-gue-Dawley健康成年雄性大鼠随机分成5组,即空白对照组、模型对照组、损伤组、RES组、甲基强的松龙(MPSS)组。根据Allen氏法制成中度脊髓损伤模型,术后立即腹腔注射RES 100 mg/kg或100 mg/kg;TUNEL法观察脊髓损伤后8,24,72 h使用RES前后脊髓神经细胞凋亡情况,并且与MPSS组进行疗效对比。结果:脊髓损伤后8,24,72 h,RES组脊髓凋亡神经细胞数与损伤组比较差异具有高度显著性(P<0.01),表明RES明显干预了损伤后脊髓神经细胞凋亡。RES组比MPSS组间的凋亡神经细胞数差异无统计学意义(P>0.05),这意味着RES可以起到与MPSS近似的效果。结论:RES能够明显抑制脊髓损伤后脊髓神经细胞的凋亡,对损伤后的脊髓有保护作用。

Abstract:Objective: To investigate the effect of resveratrol on the myelonic neuronal apoptosis in the forepart of spinal cord injury.Methods: Sixty-two adult male Sprague-Dawley rats were randomly divided into five groups including normal control,mode control,injury group,resveratrol(RES) treatment group and methylprednisolone sodium succinate(MPSS) treatment group.After the model of moderate spinal cord injury was established according to the Allen's method,resveratrol(100 mg/kg) or methylprednisolone sodium succinate(100 mg/kg) was injected immediately by intraperitioneal injection.Results: The apoptotic cells were less detected in the RES group than in the injury group at 8,24 and 72 h after spinal cord injury(all P<0.01).And the similar changes were also observed in MPSS group(P<0.05 at 8 h,and both P<0.01 at 24 and 72 h).There were no difference in apoptotic cell counts in spinal cord between the RES group and MPSS group(P>0.05).Conclusion: Resveratrol could restrain the apoptosis of the neurons after spinal cord injury.Therefore,resveratrol can protect the spinal cord in the forepart of spinal cord injury.
