脐血源性间充质干细胞向神经样细胞诱导分化的研究

摘    要:目的探讨来源于人脐带血来源的间充质干细胞(MSCs)体外诱导分化成神经样细胞的可行性。方法取第5代的脐血间充质干细胞,用不同浓度的脑源性神经营养因子(BDNF)和睫状神经营养因子(CNTF)单独或联合诱导脐源血间充质干细胞向神经样细胞分化。倒置相差显微镜观察细胞形态变化,于实验第1、3、6天分别进行神经元特异性烯醇化酶(NSE)和胶质纤维酸性蛋白(GFAP)免疫细胞化学染色并计数分化为神经元样细胞及神经胶质细胞的比例。结果与对照组相比,BDNF和CNTF能显著提高人脐血源MSCs分化为神经元的比例。其中20 ng/mL的BDNF联合20 ng/mL CNTF诱导人脐血源MSCs分化为神经样细胞的比例最高。结论人脐血MSCs经BDNF和CNTF体外诱导,能够分化为神经样细胞。

Abstract:Objective To investigate the feasibility of the induction of mesenchymal stem cells derived from human umbilical blood into neuron-like cells.Methods The human umbilical blood-derived mesenchymal stem cells were induced with different contents of brain-derived neurotrophic factor(BDNF) and ciliary neurotrophic factor(CNTF) in vitro.The concentrations of BDNF and CNTF ranged from 5 ng/mL,10 ng/mL to 20 ng/mL.The cell morphology was observed under inverted phase-contrast microscope.Specific markers,such as neuron-specific enolase(NSE) and glial fibrillary acidic protein(GFAp),were analyzed by immunocytochemistry method.Results The inducing differentiation rates of neurons were improved greatly by BDNF and CNTF compared with the control group(P<0.05).The combination of 20 ng/mL BDNF and CNTF was the most suitable for inducing differentiation of the human umbilical blood-derived mesenchymal stem cells among the experimental groups.Conclusion Human umbilical blood-derived mesenchymal stem cells are capable of differentiating into neuron-like cells induced by brain-derived neurotrophic factor(BDNF) and ciliary neurotrophic factor(CNTF) in vitro.
