杞叶含药血清对长波紫外线损伤人皮肤成纤维细胞的保护作用

摘    要:目的探讨不同浓度枸杞叶(LBL)含药血清对长波紫外线(UVA)辐射致人真皮成纤维细胞(HSF)损伤的防护作用及其作用机制。方法采用强度2.8J.cm^-2的UVA照射HSF损伤造模,实验分为空白组、对照组、UVA模型组、空白血清组、UVA+0.15g.mL^-1LBL组,UVA+1.5 g.mL^-1LBL组和VitC阳性对照组,用MTT法检测细胞增殖活性,酶生化法检测MDA含量及培养液中LDH漏出量。结果正常大鼠血清不影响HSF功能,UVA照射HSF损伤法可造成HSF增殖活性降低,MDA含量升高,LDH漏出增加,与空白组相比差异有统计学意义(P均〈0.05);不同浓度的LBL及VitC均能增加细胞的增殖活性,减少MDA含量和LDH的漏出量,但LBL作用更优(P〈0.05)。结论不同浓度LBL对UVA损伤的HSF均有一定的保护作用,其机制可能与LBL的抗氧化作用及促进细胞增殖活性有关。

Abstract:Objective To explore the protective effects and mechanism of the different concentrations of lycium barbarum leaf(LBL) containing serum on human skin fibroblast(HSF) in vitro irradiated by ultraviolet A(UVA).MethodsHSF model was established using the irradiation intensity 2.8J·cm-2 of UVA damage.Experiment were divided into blank group and control group,UVA radiated group,blank serum group,two different concentrations of LBL protective group and the VitC-positive control group.The activities of cell proliferation were measured by MTT methods.The contents of MDA and the activities of LDH in the supernatants of fibroblasts were determined by biochemical methods.ResultsNormal rat serum did not affect the function of HSF.That the fibroblasts were irradiated by UVA caused the decrease in the activities of cell proliferation and increase in the content of MDA and LDH(Compared with the blank group both P0.05).Different concentrations of LBL and the VitC could increase the activities of cell proliferation and reduce the content of MDA and LDH,but the role of better LBL.ConclusionsDifferent concentrations of LBL have protective effects on fibroblast irradiated by UVA.The mechanism of this effect may be associated with it's anti-oxidative effect and enhancing cell's proliferation.
