白脉软膏对脑缺血大鼠脑组织p38 MAPK及GAP-43蛋白表达影响的实验研究

摘    要:目的通过观察脑缺血后白脉软膏对大鼠脑组织p38MAPK及GAP-43蛋白表达的影响,探讨白脉软膏对脑组织的神经保护作用。方法60只雄性Wistar大鼠随机分为正常组、缺血组、白脉干预组,大脑中动脉线栓法制作局灶性脑缺血大鼠模型,分别于1、3、7、14d处死动物,采用免疫组织化学法观察各组脑组织p38MAPK和GAP-43蛋白的表达。结果p38MAPK缺血组的表达在7、14d分别为178.2±18.7、164.8±21.5,显著高于白脉组的153.2±21.9、120.2±18.6(P〈0.01);GAP-43白脉组的表达在7、14d分别为62.2±6.4、55.0±7.6,也显著高于缺血组的35.7±4.0、33.0±3.7(P〈0.01)。结论白脉软膏可调节脑缺血后p38MAPK和GAP-43的表达,并与损伤时间的变化有一定相关性,提示白脉软膏可能与损伤后神经元的再生、修复有关,有进一步研究的价值。

Effect of Baimai ointment on the expression of p38MAPK and GAP-43 in ischemic brain of rats

Abstract:Objective To explore the mechanism of the neuroprotection of Baimai ointment by determining the expression changes of p38MAPK and GAP - 43 in cerebral tissue after cerebral infarction in rats. Methods 60 male wistar rats were divided randomly into 3 groups: normal control group, ischemic group and Baimai ointment group. Cerebral ischemia models of rats were established by blocking the middle cerebral artery. The expression of p38MAPK and GAP - 43 in normal control group, ischemic group and Baimai ointment group were detected by immunohistochemical method on different time of 1 st, 3rd, 7th, 14th day after each management. Results The expression of Baimai Dintment in p38MAPK in ischemic group was significantly higher than Baimai ointment group ( P 〈 0.01 ) at the 7th and 14th day and GAP - 43 in Baimai ointment group was higher than ischemic group ( P 〈 0.01 ) at the 7th and 14th day. Conclusions The effect of Baimai ointment in modifying the expressions of p38MAPK and GAP - 43 may be related to time changes. Baimai ointment may play a role in the regeneration and movement of neurons.
