摘 要: | 目的探讨安宫牛黄丸对大鼠脑出血(Intracerebral Hemorrhage,ICH)后脑组织基质金属蛋白酶-9(Matrix metalloproteinase-9,MMP-9)表达的影响。方法将sD大鼠随机分为正常对照组、假手术组、脑出血组、安宫牛黄丸治疗组,每组5只大鼠。采用自体股动脉血注入尾状核建立大鼠脑出血模型,观察48h时点各组大鼠血脑屏障通透性变化,RT.PCR分析安宫牛黄丸对脑出血后MMP-9mRNA表达的影响,WesternBlot分析安宫牛黄丸对脑出血后MMP-9蛋白表达的影响。结果大鼠实验性脑出血后48hMMP-9阳性细胞数及伊文思蓝渗出量增多,MMP-9mRNA及MMP-9蛋白表达增加;安宫牛黄丸处理后,MMP-9阳性细胞数减少(P〈0.01),伊文思蓝渗出量降低(P〈0.01),MMP-9mRNA及MMP-9蛋白表达下降。结论脑出血后MMP-9表达与血脑屏障通透性密切相关,安宫牛黄丸能够有效抑制大鼠脑出血后MMP-9表达,减轻脑水肿。

Abstract: | Objective To study the effect of Angong Niuhuang Wan on MMP-9 expression after intracerebral hemorrhage (ICH)in rats. Methods In this group rebral , sham operation group, ICH group and Angong study, the SD rats were randomly divided into control Niuhuang Wan group, five rats each group. The intrace- hemorrhage model was established by injection of auto-non-anticoagulant artery blood into the caudate nucleus of the rats. 48h later, the permeability of blood brain barrier (BBB) was observed and the expression of MMP-9 mRNA and proteion were assayed by RT-PCR and Western-Blot, respectively. Results 48h after ICH in rats, the number of MMP-9-positive cells, the permeability of BBB, and the expression of MMP-9 mRNA and protein were increased as compared to sham group (P 〈 0.01 ). Compared with ICH group, Angong Niuhuang Wan, could reverse all these changes and the difference was significant (P 〈 0.01 ). Conclusion The permeability of blood brain barrier was closely related with the MMP-9 expression after ICH. Angong Niuhuang Wan can obviously inhibit the expression of MMP-9 mRNA and protein and alleviate brain tissure injury following ICH. |
