摘    要:目的:通过对肥胖哮喘动物模型发病情况、介质变化的研究,探讨肥胖在哮喘发病中的作用.方法:将雄性Wistar大鼠随机分为肥胖哮喘组(A组)、哮喘组(B组)和对照组(C组)3组各10只.A组以高脂饲料喂养,B、C组以普通饲料喂养.A、B组大鼠腹腔注射并两周后雾化吸入卵清蛋白(OVA)建立哮喘模型.取肺组织做病理切片,应用免疫组化方法检测肺组织肿瘤坏死因子-α(TNF-α)水平,ELISA双抗体夹心法测定血浆白细胞介素-8(IL-8)水平.结果:A组大鼠气道壁增厚、炎症细胞浸润均较其他两组明显,肺组织TNF-α水平(25%±3%)高于B组(16%±3%)及C组(9%±2%)(均P<0.05),血浆IL-8水平[(32.70±7.95)ng·L-1)]高于B组[(24.52±3.03)ng·L-1]及C组[(17.48±3.06) ng·L-1](均P<0.05).结论:肥胖影响哮喘发作严重程度以及体内某些哮喘相关因子的含量.

Abstract:Objective: To explore the role of obesity in asthma through animal models of obesity in asthma incidence and changes of media.Methods: The male Wistar rats were randomly divided into three groups: A.Obesity and asthma group(n=10);B.Asthma group(n=10);C.Control group(n=10).Group A was fed with a high fat diet,whereas Group B and C with normal diet.To established model of asthma,Ovalbumin(OVA) intraperitoneally injected to group A and B group to sensitize followed by inhalation of OVA two weeks later.Group C served as a control group.All the rats were sacrificed and the lung tissues were performed biopsy.Levels of TNF-α in lung tissues were detected with immunohistochemistry.Interleukin-8(IL-8)levels in plasma were measured by enzyme linked immunosorbent assay(ELISA).Results: In group A the airway wall thickening,inflammatory cell infiltration was significantly higher than those of other groups;levels of TNF-α in lung tissues(25% ±3%) were higher than those of other groups[(16% ±3%),(9% ±2%)](P<0.05).IL-8 levels in plasma[(32.70±7.95)ng·L-1] were higher than those of other groups[(24.52 ±3.03),(17.48±3.06)ng·L-1](P<0.05).Conclusion: Obesity affects the severity of asthma attacks and some of asthma-related factors.
