摘    要:目的:利用响应面分析法优化锁阳黄酮超声提取工艺.方法:在单因素实验基础上,根据Box-Benhnken中心组合设计原理,以乙醇浓度、料液比、提取温度、超声提取时间为自变量,总黄酮提取率为响应值,设计4因素3水平响应面分析试验,来确定锁阳总黄酮最佳超声提取工艺.结果:锁阳总黄酮最佳提取工艺为:超声功率为600W、提取2次条件下,以乙醇浓度为70%;料液比1:30;提取温度77℃;超声提取时间62min,锁阳总黄酮提取率达28.815%.结论:响应面分析所得二次模型方程能较好地预测实验结果,实验设计和响应面法优化得到的提取工艺参数准确可靠,该研究可为锁阳资源的合理开发和质量控制提供理论依据和数据参考.

Abstract:Objective:Response surface analysis methodology was applied to optimize the ultrasonic-assisted extraction conditions of total flavonoids from Cynomorium songaricum Rupr.Method:Based on the single-factor experiments,a central composite design was used to investigate the effects of four independent variables,namely ethanol concentration,the solid-liquid ratio,ultrasonic extraction temperature and ultrasonic extraction time on the response,yield of flavonoids.Results:The optimum ultrasonic extraction conditions were:extracting two times,ultrasonic power 600W,ethanol concentration 70%,the solid-liquid ratio 1:30,extraction temperature 77℃,extraction time 62min under the optimum conditions,the theoretical value of extraction yield of flavonoids was up to 28.815%.Conclusion:Response surface method for optimization of the extraction process is accurate and reliable.This study could provide theoretical basis and reference data for rational development and quality control of Cynomorium songadcum Rupr.
