细胞周期相关基因的表达与壶腹癌细胞增殖关系的研究

摘    要:目的探讨细胞周期调控相关基因p53、p21、p16和Rb与壶腹癌细胞增殖之间的关系及细胞增殖相关蛋白Ki-67与壶腹癌生物学特性和预后之间的关系。方法采用免疫组化ABC法对40例壶腹癌和10例正常壶腹组织进行染色。结果4种抑癌基因在壶腹癌中的阳性表达率分别为50%、62.5%、47.5%和92.5%。p21、p16的表达与Ki-67指数呈负相关(P<0.05),p53、Rb与Ki-67指数无相关性(P>0.05),而Ki-67指数与壶腹癌的病理分级、临床分期明显相关(P<0.05)。结论p21,p16异常表达与壶腹癌细胞异常增殖有关。Ki-67指数可能有助于了解壶腹癌的生物学特性并对预后进行判断。

Abstract:Objective To study the relationship between the cell cycle-related genes p53, p21, p16, Rband proliferation of ampullary carcinoma cells, and relationship between the cell proliferation-related Ki-67 proteinand biological property and prognosis of ampullary carcinam. Method The expression of these 4 genes and Ki-67protein in tissues of normal ampullas (n=10) and of ampullary carcinoma(n=40) was detected by immunohistochemical ABC method. Results The positive rates of p53, p21, p16 and Rb proteins in ampullary carcinomawere 50%, 62.5%, 47.5% and 92.5%, respectively. The correlation between the expression of p21, p16 proteins and Ki-67 index wes negative. No association o f p53, Rb expressions with Ki-67 index wes found. There wereclose relationships between the Ki-67 index and histological grades, clinical stages. Conclusion Abnormal expession of p21, p16 proteins is associated with tumor cell proliferation and the detection of Ki-67 index may beuseful in understanding biological characteristics as well as evaluating prognosis in ampullary carcinoma.
