胃癌细胞bcl-2和Caspase3的表达对TRAIL抗瘤作用的影响

摘    要:目的 探讨胃癌细胞bcl-2和Caspase3的表达对TRAIL抗瘤作用的影响。方法 应用细胞培养、P1染色、流式细胞仪检测的方法,观察胃癌细胞SGC-7901、MKN28、AGS、MKN45经TRAIL作用后细胞周期及细胞凋亡率,western blot方法分析4种胃癌细胞中bel-2和Caspase3的表达。结果 TRAIL300ng/ml作用于胃癌细胞24h后,胃癌细胞MKN28、MKN45、AGS和SGC-790I的凋亡率分别为36.05%、20.27%、16.50%和11.80%。Western blot结果显示MKN28细胞Caspase3的表达高于其它细胞,而bel-2的表达在各细胞间并无明显差别。结论 胃癌细胞对TRAIL的敏感性与Caspase3的高表达有关,而与bel-2的表达无关。

Abstract:Objective To study the effects of expression of bcl 2 and Caspase3 on TRAIL anti tumor activity in gastric cancer cells.Methods Gastric cancer cells were cultured in RPMI 1640 and the cell cycles and the percent of apoptosis were analysized by flow cytometer. The expressions of Caspase3 and bcl 2 in gastric cancer cell were evaluated by western blot. Results After gastric cancer cells were exposed to TRAIL 300ng/ml for 24 hours, the apoptosis percent was 36.05% for MKN28, 20.27% for MKN45, 16.50% for AGS and 11.80% for SGC 7901. Western blot showed the expression of Caspase3 was higher in MKN28 than in MKN45, AGS and SGC 7901. The expression of bcl 2 had no difference among the four cell lines. Conclusions The anti tumor activity of TRAIL is related to the expression of Caspase 3,but not to the expression of bcl 2.
