染料木黄酮联合5-FU对结肠癌HCT-116细胞株增殖及凋亡的影响

摘    要:目的探讨染料木黄酮联合5-氟尿嘧啶对HCT-116细胞增殖和凋亡的影响及其可能的作用机制。方法经不同浓度的Gen和(或)5-FU处理HCT-116细胞后,用MTT法检测细胞的生长抑制效应;流式细胞术检测细胞凋亡和细胞周期变化情况;RT-PCR检测Suvivin表达水平。结果 Gen与5-FU联合应用后能明显抑制细胞增殖,诱导细胞凋亡,使细胞发生S期阻滞,均较对照组和单用药组作用增强,并且下调了Suvivin的mRNA表达,差异具有统计学意义(P<0.05)。结论 Gen可抑制结肠癌HCT-116细胞的增殖,将细胞阻滞于S期,并诱导细胞的凋亡,与5-FU联合应用具有显著协同效应,其机制可能与下调Suvivin表达有关。

The Effect of Genisterin Combined with 5-FU on the Proliferation and Apoptosis of Human Colorectal Carcinoma Cell Line

Abstract:Objective To investigate the effect of genisterin (Gen) combined with 5-Fluorouracil (5-FU) on the proliferation and apoptosis of human colon carcinoma HCT-116 cells and the possible mechanism. Methods HCT-116 cells were treated with Gen and 5-FU of difference concentrations and MTT assay was applied in detecting the cell growth inhibition effect; cell cycle and apoptosis rate was examined by flow-cytometry (FCM) and the expression of Survivin was detected by RT-PCR, Results Gen combined with 5-FU obviously inhibited the proliferation of the cells and induced the apoptosis and the S phase arrest, the effect was much stronger than those in control group and single-drug group and down-regulated the mRNA expression of Survivin, the difference was of statistical significance (P〈0.05). Conclusions Gen can inhibit the proliferation of human colon carcinoma HCT-116 cells and induce the apoptosis and S phase arrest, obvious synergistic effect could be gained when combined with 5-FU, the mechanism might be associated with the down-regulation of Survivin expression.
