| 摘 要： | 目的 采用AdEasy系统构建的带有标记基因RFP的重组腺病毒Ad-Cyr61感染人肝癌细胞株HepG2,并观察外源性cyr61对肿瘤细胞生长和迁移的影响.方法 Ad-Cyr61与Ad-RFP分别感染人肝癌细胞株HepG2,荧光倒置显微镜观察荧光表达,通过RT-PCR、免疫组化实验分别检测Ad-Cyr61的mRNA和蛋白的表达;通过M1Tr实验、细胞计数实验、克隆形成实验和划痕愈合实验,观察Cyr61对人肝癌细胞株HepG2细胞增殖和迁移的影响.结果 Ad-Cyt61感染人肝癌细胞株HepG2,与对照组比较细胞数量增多,集落形成增加,细胞划痕逐渐愈合.结论 Ad-Cyr61感染人肝癌细胞株HepG2后可促进细胞增殖和迁移,提示Cyr61在肿瘤发生和转移中可能起到重要作用.
