摘    要:PEG-PLA/PLGA/PCL聚合物药物载体因具备可生物降解性、高包封率、靶向运输、可控释放及可修饰的特性,在癌症治疗的应用中得到越来越广泛的关注。然而,PEG-PLA/PLGA/PCL聚合物材料作为抗癌药物的递送载体仍然面临诸多的挑战:1)聚合物药物载体结构的不稳定性;2)靶向运输的效果不明显;3)药物在肿瘤组织释放不完全等。因此,设计出新型的PEG-聚酯类聚合物药物载体成为研究的前沿和热点。本文综述了近年来国内外研究学者在构建多功能性的PEG-聚酯类聚合物纳米抗癌药物载体的研究成果,从提高结构稳定、增强靶向能力以及刺激响应释放三种设计理念对不同的纳米载药体系进行讨论、分析和总结。

Abstract:The application of PEG-PLA/PLGA/PCL polymer as novel drug delivery system for cancer treatment has been extensively investigated due to its biodegradability, high encapsulation efficiency, targeted transportation and controllable release profile of drug, tunability of structure and properties. However, there are still many challenges in the development of PEG-PLA/PLGA/PCL polymeric anticancer drug carriers: 1) structural instability of polymeric drug carriers; 2) poor effect of targeted transportation; 3) insufficient drug release in tumor tissue, etc. Therefore, the design of novel PEG-polyester polymeric drug carriers has been a hot area of research. This paper reviewed the study of new designs of multi-functional biodegradable PEG-PLA/PLGA/PCL polymeric nano-anticancer drug carriers in recent years, and a series of different nano-carrier systems will be discussed in three design concepts: improving structural stability, enhancing targeting ability and stimulating drug release.
