新型化合物F-2对人肺癌NCI-H520细胞增殖、迁移及凋亡的影响

摘    要:目的:探讨新型化合物F-2对人非小细胞肺癌NCI-H520细胞增殖迁移及凋亡的影响。方法: CCK-8实验检测肺癌细胞增殖情况;采用划痕实验检测化合物对H520细胞迁移的情况;倒置显微镜观察化合物刺激对细胞形态的影响;流式细胞术实验检测化合物对细胞凋亡的影响;Western blot实验检测化合物对细胞迁移及凋亡相关蛋白表达的影响。结果:化合物对H520细胞增殖有明显的抑制作用,且与浓度和时间呈依赖性关系。划痕实验结果表明化合物能够明显抑制肿瘤细胞迁移,并且western blot 实验结果表明化合物处理H520细胞后可上调E-cadherin的表达,下调N-cadherin蛋白的表达。Annexin-V/PI双染法结果显示,化合物可诱导H520细胞发生凋亡,且随着药物浓度增加凋亡率上升,并且增加了BaX/BcL-2蛋白表达比率。结论:化合物能够明显抑制肺癌细胞的增殖、迁移及诱导细胞凋亡,可能是通过阻断肿瘤细胞EMT过程,调节细胞凋亡过程中相关基因表达从而发挥抗肿瘤作用。

Abstract:ABSTRACT: OBJECTIVE To investigate the effects of novel compound F-2 on proliferation, migration and apoptosis of human non-small cell lung cancer NCI-H520 cells. METHODS The proliferation of lung cancer cells was detected by CCK-8 assay. The migration of H520 cells was detected by wound scratch assay. The effect of compound stimulation on cell morphology was observed by inverted microscope. The effect of compounds on apoptosis was detected by flow cytometry. The effects of compounds on cell migration and apoptosis-related protein expression were detected by Western blot assay. RESULTS The compounds significantly inhibited the proliferation of H520 cells in a dose and time dependent manner. The wound scratch assay showed that the compound could significantly inhibit the migration of tumor cells, Western blot results showed that the compound could up-regulate the expression of E-cadherin and down-regulate the expression of N-cadherin in H520 cells. Annexin-V/PI double staining showed that the compound could induce apoptosis of H520 cells, and the apoptotic rate increased with the increase of drug concentration, and the expression ratio of BaX/BcL-2 protein increased. CONCLUSION Compounds can significantly inhibit the proliferation, migration and induce apoptosis of lung cancer cells, which may be through blocking the EMT process of tumor cells and regulating the expression of related genes during apoptosis.
