复合骨形态发生蛋白2及碱性成纤维细胞生长因子注射型骨修复材料对兔骨髓基质细胞增殖和超微结构的影响

摘    要:目的观察以纤维蛋白胶(fibrinsealant,FS)为载体的注射型骨修复材料对体外培养的兔骨髓基质细胞(marrowstromalcell,MSC)增殖和超微结构的影响。方法实验分组为实验组(FS+bFGF+rhBMP-2)、对照组1(FS)、对照组2(FS+bFGF)、对照组3(FS+rhBMP-2)及单纯对照组。采用细胞培养、MTT及电镜等方法对各组兔MSC的增殖及超微结构进行研究。结果(1)各组材料对细胞促增殖作用由强到弱依次是:对照组2(FS+bFGF)、实验组(FS+bFGF+rhBMP-2)、对照组1(FS)、对照组3(FS+rhBMP-2)、单纯对照组,差异有显著性(P<0.05);(2)扫描电镜观察发现,材料表面粗糙,有微孔存在,各组细胞与材料融合生长。透射电镜观察发现:实验组的绝大部分细胞呈成骨细胞表型,细胞增殖状态旺盛,分化好。各对照组细胞或是细胞增殖活性高,但向成骨细胞表型分化差,或是向成骨细胞表型分化好,但细胞增殖活性较差。结论FS+bFGF+rhBMP-2即可显著促进MSC增殖和兔MSC向成骨细胞方向的分化水平。

Abstract:Objective To investigate the influence of the injectable osteoinductive material with rhBMP 2 and bFGF on the proliferation and ultrastructure of marrow stromal cell MSC of rabbit.Method We investigate the influence of experiment group(FS+bFGF+rhBMP 2),control group 1(FS),control group 2(FS+bFGF),control group 3 (FS+rhBMP 2) and Single control group (no any material ) on the proliferation and the ultrastructure of rabbit MSC by electron microscopy and cell culture.Result (1)The proliferation rate of rabbit MSC of experiment group(FS+bFGF+rhBMP 2) was significantly higher than control group 1(FS), control group 3 (FS+rhBMP 2) and Single control group (no material) ,but lower than control group 2(FS+bFGF) (P< 0.05).(2)Investigation under scanning electron microscope found that the surface of material was rough and had many pores and that cell and material mixed and grew together.Investigation under transmission electron microscope found that MSC of experiment group were mostly phenotype of osteoblast with high proliferation activity and high differentiation degree toward osteoblast MSC of control groups were either low differentiation degree toward osteoblast with few extracellular matrix and collagen fibers or low proliferation activity.Conclusion The injectable osteoinductive Material with FS+bFGF+rhBMP 2 could significantly accelerate the proliferation and differentiation towards osteoblasts of marrow stromal cell MSC of rabbit.
