利用胶体金免疫层析技术快速定量检测单增李斯特菌方法的建立

〔目的〕建立胶体金免疫层析技术快速定量检测单增李斯特菌方法。〔方法〕利用胶体金标记和双抗体夹心免疫层析技术,建立单增李斯特菌的快速检测方法,评价其特异性和敏感性,并拟合标准曲线进行定量检测;在牛奶、橙汁、果冻等即食食品样品中添加单增李斯特菌模拟污染样品,评价该方法对半固体、液体等即食食品、可疑生物恐怖样品的检测能力。〔结果〕该法可在10min内完成定性和半定量检测,定量检测灵敏度为3.5×103cfu/ml,线性范围3.5×105～3.5×108cfu/ml,回收率在99%～101.7%之间。〔结论〕建立的检测单增李斯特菌的胶体金免疫层析方法,能快速、灵敏、特异、准确地对样品中的单增李斯特菌进行定性、定量检测。

Rapid and Quantitative Detection of Listeria monocytogenes Using Colloid Gold Immunochromatographic Method

Objective To develop a rapid and quantitative detection method on Listeria monocytogenes using colloid gold immunochromatographic method.Methods The rapid detection method was established with double-antibody sandwich assay,and the specificity and sensitivity of the method was then evaluated.Quantification was realized by constructing a standard curve,Milk powder,milk solution and ham sausage,added with ricn were tested to evaluate the feasibility on solid,semi-solid,liquid food samples and doubtful bioterrorism samples by this method.Results This established method could be accomplished qualitatively as well as semi-quantitative detection in 15 minutes,the sensitivity was up to 3.5×103 cfu/ml,the linear range was from 3.5×105cfu/ml to 3.5× 108cfu/ml,the recovery rate was 99%～101.7%.This method showed good specificity and sensitivity,which could detect samples directly.Conclusion The colloidal gold immunochromatographic test,which could detect the abrin in samples rapidly,specifically,sensitively and accurately,was able to realize quantification,and is suitable for field detection.
