| 摘 要： | 目的 在体外贴壁诱导E14小鼠胚胎干细胞分化为甲状腺细胞的基础上,不改变诱导因子,探索鼠尾胶原三维结构体外诱导胚胎干细胞分化为甲状腺细胞的可行性.方法 E14小鼠胚胎干细胞半固体培养基悬浮培养6 d生成拟胚体,随后在鼠尾胶原三维结构中继续诱导分化11d,全程添加诱导因子促甲状腺素及胰岛素,第14天时开始添加碘化钾.结果 拟胚体在鼠尾胶原上生长状态良好,分化细胞贴壁后逐渐陷入鼠尾胶原;电镜下诱导细胞高尔基体、内质网丰富,部分诱导细胞胞质内见不典型分泌颗粒,与人成体甲状腺细胞超微结构类似.结论 胚胎干细胞在鼠尾胶原三维结构中可以分化为甲状腺类似细胞,并具备合成、分泌的功能;三维结构为体外诱导胚胎干细胞分化为甲状腺细胞提供了新的方法. |
