P21在肾小球系膜细胞的表达变化及意义

摘    要:目的:探讨高糖、胰岛素对肾小球系膜细胞中细胞周期蛋白依赖性激酶抑制剂P21表达的影响及其与系膜细胞肥大的关系。方法:将培养的大鼠1097系膜细胞株分为8组:正常对照组,高糖组,甘露醇组,不同浓度胰岛素组,高糖加不同浓度胰岛素组。用RT-PCR法检测各组系膜细胞P21mRNA表达。流式细胞仪定量检测各组系膜细胞前向角度散射光(FSC)大小。结果:正常对照组系膜细胞中P21mRNA有一定表达。高糖组P21mRNA表达明显增加为正常对照组的3.89倍(P<0.01);FSC为正常对照组的1.42倍(P<0.01)。不同浓度胰岛素组系膜细胞P21mRNA表达及FSC大小随胰岛素浓度增加而增加,但无统计学意义(P>0.05)。结论:(1)P21mRNA在正常肾小球系膜细胞有一定表达。(2)高糖刺激可使系膜细胞P21mRNA表达上调,且与渗透压无明显关系。(3)P21mRNA表达上调可导致系膜细胞肥大,在糖尿病肾病发病机制中起重要作用。

Abstract:Objective:Recent studies have shown that cyclin kinase inhibitor P_(21) play an important role in diabetic nephropathy(DN).In order to explore mechanism of diabetic nephropathy and effects of P_(21),we investigated the expressive changes of P_(21) messenger ribonucleic acids(mRNA) levels in cultured rat glomerular mesangial cells.Methods:Cultured 1097 rat glomerular mesangial cells were divided into 8 groups:High glucose-treated group;Mannitol-treated group;Insulin-treated groups along with different concentration;High glucose and Insulin-treated group along with different concentration and control.Semi-quantity RT-PCR method and Flow cytometry were used to detect P_(21)mRNA expression and GMC volumes.Results:Control group has the expression of P_(21)mRNA.High glucose could increase the expression of P_(21)mRNA to 389% and the value of forward scatter(FSC) 142% versus control(P<0.01).There was no significant difference in insulin-treated group.Conclusion:1.The expression P_(21)mRNA exists in normal glomerular mesangial cells.2.High glucose may increase significantly expression of P_(21)mRNA in mesangial cells and lead to cells hypertrophy.3.P_(21) has an important role in the pathogenesis of DN.
