survivin在非小细胞肺癌中表达的意义及其与caspase-3、DFF45蛋白之间的相互关系

目的探讨survivin蛋白的表达在非小细胞肺癌(NSCLC)发生、发展方面的作用及其与临床病理特征之间的联系,与caspase3及DFF45表达的相互关系。方法运用免疫组织化学SP法对60例NSCLC、10例淋巴结转移癌、10例正常支气管粘膜上皮石蜡切片组织中survivin、caspase3及DFF45蛋白的表达进行检测。结果NSCLC、淋巴结转移癌中survivin的阳性表达率分别为70%及90%,显著高于正常支气管粘膜上皮的阳性率10%(P<0.01)。survivin在NSCLC中的表达与分化程度(P<0.01)、TNM分期(P<0.05)、淋巴结转移(P<0.05)有关。而与年龄、性别、病理类型无明显相关(P>0.05)。survivin表达与caspase3的表达呈负相关(P<0.01;r=-0.335),而与DFF45的表达却呈正相关(P<0.01;r=0.406)。结论survivin在NSCLC的发生、发展中起重要作用,可作为NSCLC的诊断指标,对NSCLC恶性程度的判断及预后也有一定参考价值。survivin的作用是通过抑制caspase3的表达,从而影响下游凋亡途径而发挥作用的。

Expression of survivin Protein and Its Relationship with Caspase-3, DFF45 Protein in Non-small Cell Lung Cancer

Objective To investigate the expression of survivin protein in non-small cell lung cancer (NSCLC)and the clinicalpathological characteristic of lung cancer, to evaluate the relationship between expression of survivin,caspase-3 and DFF45 proteins. Methods The expression of surviving and caspase-3, and DFF45 proteins was evaluated by immunohistochemistry in 10 normal bronchial epithelium, and 60 NSCLC and 10 metastatic carcinoma of lymph node.Results The expression of survivin protein was detected in a significantly greater proportion in NSCLC(70%) and metastatic carcinoma of lymph node(90%) than normal bronchial epithelium(10%). The up-regulation expression of survivin was related to the TNM stages(P< 0.05), cell differentiation(P< 0.01) and lymph node metastases(P< 0.05), but no relation with age, sex and histological subtype(P> 0.05). Expression of caspase-3 was negatively(P< 0.01;r= -0.335) and DFF45 positively correlated with expression of survivin(P< 0.01;r= 0.406). Conclusion survivin may play an important role in the progression of NSCLC and be identified as a defining diagnostic marker. survivin is worthy of analyzing malignant grade and judging the prognosis of NSCLC.Survivin can resist cell apoptosis by restrainting caspase-3 expression.
