铅对孕鼠生殖内分泌影响的研究

摘    要:应用放射免疫分析法(RIA)和放射配体受体法研究铅对孕鼠生殖内分泌和子宫雌激素受体的影响。SD雌性大鼠于妊娠第7~12天,每天灌服120mg/kg和220mg/kg硝酸铅水溶液。孕鼠体重、胚胎重量、子宫重量和活胎数与对照组无显著差异。高剂量组孕鼠血清黄体生成激素(LH)、生乳素(PRL)和孕酮(P)含量明显高于对照组(P<0.01);染毒组血清促卵泡激素(FSH)和雌二醇(E_2)含量与对照组无显著差异。染毒组孕鼠子宫E2受体数与对照组无显著差异,但高剂量组受体亲和力(Ka)明显高于对照组(P<0.01)。孕鼠垂体促性腺激素分泌细胞超微结构无明显病理改变。认为铅可能干扰下丘脑-垂体-卵巢轴的调控机制,导致生殖内分泌功能紊乱。

Abstract:The effect of lead on reproductive endocrine function and estrogen receptors in uteri of pregnant rats were analyzed by radioimmunoassay and radioligand receptor assay. Female SD rats were orally administered daily with 120mg/kg and 220mg/kg lead nitrate solution from 7th to 12th day of pregnancy. There was no difference in body weight , embryo weight,uterus weight and survival emb ryo number between treated and control groups.The contents of luteinizing hormone(LH),prolactin (PRL) and progesterone(P) were significantly higher (P<0.01) in high dose group while follicle stimulating hormone(FSH)and estradiol (E_2) were not elevated remarkably in treated groups compared with control group. Treated group did not obviously differ from control in the number of estradiol receptors,but affinity of receptors in high dose group increased(P<0. 01) with reference to control.In addition,there appeared no notable pathologic changes in ultrastructure of secretary cells for gonadctropic hormone and prolactin in rat pituitary.It is concluded that lead may interfere with the regulatery mechanism of hypothalamic-pituitary-ovarian axis to disturb reproductive endocrine function of pregnant rats.
