前列腺素E1对乳酸盐腹膜透析液体外致人腹膜问皮细胞损伤的保护作用

摘    要:目的 传统乳酸盐腹膜透析液(L—PDS)对人腹膜间皮细胞(HPMC)具有细胞毒作用。本研究旨在观察前列腺素E1(PGE1)对L—PDS在体外致人HPMC活力和增殖抑制的影响。方法 采用胰蛋白酶消化法培养HPMC;设立培养基对照组、L—PINS组及PGE1组。用乳酸脱氢酶(LDH)的释放和四唑盐比色法(MTT法)评估细胞活力及检测细胞增殖。结果 L—PDS可使HPMC的LDH释放显著增多(P〈0.01)。随暴露于L—PDS时间延长,存活率进行性下降。PGE1可减少LDH释放而提高细胞存活率(P〈0.05)。L—PDS作用30min能抑制HPMC增殖(P〈0,05),而PGE1能促进其增殖(P〈0.05)。结论 PGE1对L—PDS体外所致HPMC活力和细胞增殖的抑制有一定的保护作用。

Abstract:Objective Lactate-based peritoneal dialysis solutions (L-PDS) showed cytotoxic effect to human peritoneal mesothelial cells (HPMC). The aim of this research was to observe the influence of prostaglandin E1 (PGE1) on viability and proliferation of HPMC, which were inhibited by L-PDS in vitro. Methods Mesothelial cells were cultured after trypsin digestion. The cultured HPMC were divided into control group, L-PDS group, and PGE1group. The viability of HPMC was assessed by lactate dehydrogenase (LDH) release and tetrazolium salt colorimetry assay (MTT assay); MTT assay was also used to assess the growth of the cells. Results L-PDS increased LDH release as compared with the control (P< 0.01 ). When the HPMC exposed to L-PDS prolonged, the viability of cells decreased progressively. Treatment with PGE1 reduced the release of LDH, increased the cell viability (P<0.05). Otherwise, exposed to L-PDS for 30 minutes, the proliferation of HPMC was inhibited (P<0.05). PGE1 enhanced the proliferation of HPMC (P<0.05). Conclusion PGE1 had some protective effects on cell viability and proliferation of HPMC, which were inhibited by L-PDS in vitro.
