小肠三叶因子对盐酸诱导的大鼠胃溃疡的预防和治疗作用

摘    要:目的 探讨克隆表达的rh ITF在模拟胃肠道条件下的稳定性和抗胃溃疡的作用机制。方法 采用质谱法及盐酸诱导的大鼠胃溃疡模型 ,测定rh ITF在模拟胃肠道条件下的结构稳定性和大鼠腺胃粘膜损伤级别 ,损伤指数及胃酸度 ,胃蛋白酶活性及胃排空能力。结果 酸对rh ITF相对分子量没有影响 ,胃蛋白酶仅从N端切除 6个氨基酸残基。rh ITF可明显降低大鼠腺胃粘膜损伤级别和损伤指数 ,但对胃酸分泌 ,胃蛋白酶活性及胃排空能力没有影响。结论 rh ITF在体外对酸和胃蛋白酶有一定的抗性 ,其对胃溃疡的治疗和预防作用不是通过降低攻击因子 (酸和蛋白酶 )和影响胃的自主运动实现的。

Abstract:AIM To study the protective and therapeutic mechanism of rh ITF against hydrochloric acid induced gastric ulcer in rats and its structural stability under the mimic condition of gastrointestinal tract. METHOD MALDITOF MS and hydrochloric acid induced gastric ulcer in rats were used to examine the structural stability and the grade and index of ulcer, structural stability and the gastric acid secretion, pepsin activity and gastric peristalsis ability. RESULTS Acid had no effect on the molecularweightofrh ITFandpepsinonlycutsixN terminal aminoacidresiduesofrh ITF .rh ITFcouldobvious lyreducethegradeandindexofulcer (P <0 0 5orP <0 0 1)withoutanyeffectsonthegastricacidsecre tion ,pepsinactivityandgastricperistalsisabilityby injecting 2 0 0 μg ,50 0 μg ,10 0 0 μg ,rh ITF·rat- 1 intoitsveinoftail.CONCLUSION rh ITFissome whatresistanttodegradationofacidandproteinase anditsprotectiveandtherapeuticfunctiononulceris notrelatedtoreducingattractivefactorssuchasacid andpepsinandautomobilityofstomach .Itmightbe relatedtoitsfunctionofstimulatingcellmigration .
