上皮性卵巢癌患者外周血CD4~+CD25~(high)调节性T细胞分析

目的探讨CD4+CD25highT细胞(Treg)在60例上皮性卵巢癌患者外周血中的分布及其表型特征,分析手术对卵巢癌患者外周血Treg的影响。方法采用流式细胞术检测卵巢癌患者及健康人外周血中Treg的比例,探讨其表型特征并分析手术对患者外周血Treg比例的影响。结果与正常对照(2.95±1.09)%相比,60例上皮性卵巢癌患者外周血中Treg比例明显增加(9.81±5.19)%,二者差异有统计学意义(P0.01);手术后,患者外周血中Treg的比例相比术前显著下降(P0.01);Treg表型分析发现:Treg高表达CD45RO、HLA-Ⅰ、Foxp3分子,较高水平表达OX40、GITR、CD152、CD95、CD95L、B7-H1,几乎不表达CD80、CD86、B7-H4、CD45RA、CD69分子,而HLA-DR、CD154的表达水平在个体间差异较大,同时大部分Treg细胞CD127表达阴性。结论上皮性卵巢癌患者外周血CD4+CD25highT细胞水平明显升高,可能参与卵巢癌患者肿瘤免疫抑制。

Prevalence of CD4+ CD25high Regulatory T Cells in Peripheral Circulation of Patients with Epithelial Ovarian Cancer

Objective To investigate the proportion and the phenotype of CD4^＋CD25^high T cell in the peripheral blood of 60 patients with epithelial ovarian cancer.Methods Flow cytometry was used to evaluate the prevalence of CD4^＋CD25^high T cells （Tregs） in the peripheral blood of 60 patients with epithelial ovarian cancer and 14 healthy controls.We also investigated the change of the frequency of Tregs in the peripheral blood of 10 patients before and after operation.Moreover,the phenotype of Tregs was determined by flow cytometry too.Results Compared with normal controls（2.95±1.09）%,the prevalence of Treg in the peripheral blood of 60 patients with epithelial ovarian cancer was significantly increased （9.81±5.19）%,and the difference was statistically significant （P0.01）.After the operation,the frequency of Tregs was decreased significantly （P0.01）.Additionally,we found that the Tregs had high expression of CD45RO combined with low expression of CD45RA and CD69.The costimulatory molecules CD80,CD86,and B7-H4 could rarely be detected on the surface of Treg cells,whereas Foxp3 and HLA-Ⅰwere highly expressed.OX40,GITR,CD152,CD95,CD95L,and B7-H1 were expressed on the surface of Treg cells at relatively high levels.Some of the Treg cells had variable expression of HLA-DR and CD154.Moreover,most Treg cells were negative for CD127 expression.Conclusions The prevalence of Treg cells was significantly increased in the peripheral blood of patients with epithelial ovarian cancer.These cells may negatively regulate antitumor immune response of the patients.
