摘    要:目的:观察吸附有骨形态发生蛋白(BMP)及碱性成纤维细胞生长因子(bFGF)的双相陶瓷生物骨(BCBB)植入兔股部肌袋内的成骨过程,评价BCBB/BMP/bFGF的成骨作用.方法:成年新西兰大白兔60只,随机分为4组;A组股部肌袋内植入BCBB/BMP/bFGF,B组植入BCBB/BMP,C组植入BCBB/bFGF,D组植入BCBB;分别于术后4、8和12周取材,每个时间点每组取5例标本进行外观及组织切片观察,并检测钙含量及钙磷比.结果:外观见各时间点标本A组较B组、C组和D组色泽红润,与周围组织连接紧密,组织切片术后各时间点新骨形成面积比较,A组大于B组、C组和D组,B组大于C组和D组,差异具有统计学意义(P<0.05),C组、D组间差异无统计学意义(P>0.05);X射线能谱分析,术后12周时A组钙含量及钙磷比大于B组、C组和D组,差异具有统计学意义(P<0.05).结论:BCBB是生长因子的良好吸附载体,且吸附有BMP及bFGF的BCBB具有较强的成骨能力,bFGF能够促进BMP的诱导成骨作用.

Osteogenic Effect of Tissue Engineering Bong Composed with BCBB,BMP and bFGF

Abstract:Objective: To observe the osteogenesis of biphasic ceramic biologic bone (BCBB) ab- sorbing bone morphogenetic protein (BMP) and basic fibroblast growth factor (bFGF) implanted into muscle bag of rabbit thigh, and to evaluate the osteogenic effect of BCBB/BMP/bFGF. Methods: Six- ty rabbits were randomly divided into four groups:group A, B, C and D implanted with BCBB/BMP/ bFGF, BCBB/BMP, BCBB/bFGF and BCBB into muscle bag of rabbit thigh respectively. The speci- mens were harvested separately in 4, 8 and 12 weeks after operation. 5 samples were taken at each time point in each group. Appearance and histological observation were carried out, the calcium con- tent and the ratio of calcium and phosphorus were detected. Results: The appearance of the samples in group A was redder than those of groups B, C and D, and closely connected with surrounding tissue. Histological observation found that new bone areas of the samples in group A were larger than those of groups B, C and D, and those of group B were larger than in groups C and D ( P 〈 0.05 ), and the difference between group C and D was not significant (P 〉 0.05 ). X-ray energy spectrum analysis (EDAX) showed that the calcium content was more and the ratio of calcium and phosphorus of group A was higher than those of group B, C and D in 12 week after operation, and the differences were obvi- ous(P 〈 0.05). Conclusions: BCBB is a fine absorption carrier of growth factors, and BCBB ab- sorbed BMP and bFGF has strong osteogenic activity, and the osteogenesis effect of BMP can be pro-rooted by bFGF.
