摘    要:目的:建立超高效液相色谱法测定酸水解前后化橘红中柚皮苷、野漆树苷、柚皮素、芹菜素的含量。方法:分别以超声法和酸水解法提取样品,超高效液相色谱法检测。采用WATERS ACQUITY UPLC色谱系统,ACQUITY BEH C18色谱柱(2.1mm×50mm,1.7μm),以甲醇-0.1%乙酸水溶液为流动相梯度洗脱,流速0.5mL/min,柱温30℃,检测波长283nm,289nm,337nm。结果:酸水解前可同时测定柚皮苷、野漆树苷、柚皮素、芹菜素的含量,酸水解后柚皮苷、野漆树苷分别转化为柚皮素、芹菜素,4种黄酮与其它成分均达到良好的分离。结论:该方法高效、快速、灵敏,为化橘红的抗氧化活性研究及其新制剂的开发提供参考。

Abstract:Objective: To develop the UPLC method for determination of naringin,rhoifolin,naringenin and apigenin in Exocarpium Citrus grandis before and after acid hydrolysis.Methods: The samples were extracted by ultrasonic and acid hydrolysis.Quantification was performed by UPLC at 30℃ using ACQUITY UPLC BEH C18 column(2.1mm×50mm,1.7μm),methanol-0.1% acetic acid was applied as mobile phase and DAD detector was set at 283nm,28nm,337nm.The flow rate was 0.5 mL/min.Results: The four flavonoids can be detected before acid hydrolysis.After acid hydrolysis,naringin and rhoifolin translated into naringenin and apigenin,the result showed naringin,rhoifolin,naringenin and apigenin had good separation with other components.Conclusion: This method is efficient,quickly,sensitive and provides a reliable reference to antioxidation research and exploit new preparation for Citrus grandis.
