诃子抗大鼠溃疡性结肠炎免疫机制实验研究

摘    要:目的:观察单味诃子水煎剂对大鼠溃疡性结肠炎的治疗作用并研究其抗溃疡性结肠炎的免疫作用机制。方法:Wistar大鼠32只,随机分为空白对照组、模型组、诃子水煎剂组、SASP对照组,除空白对照组外,均用6%乙酸制备溃疡性结肠炎大鼠模型。造模成功后,诃子水煎剂组和SASP对照组分别给予单味诃子水煎剂和柳氮磺吡啶肠溶片(SASP)灌胃治疗7d,同时空白对照组和模型组给予等量的生理盐水。应用免疫组化法对大鼠结肠黏膜组织中肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)的表达进行测定。结果:与空白对照组相比,模型组大鼠结肠黏膜组织中TNF-α、IL-6的表达水平显著升高(P0.01);与模型组相比,诃子水煎剂组大鼠结肠黏膜组织中TNF-α的表达水平显著降低(P0.01),IL-6的表达水平显著降低(P0.05)。结论:单味诃子水煎剂对大鼠溃疡性结肠炎有一定治疗作用,其治疗溃疡性结肠炎的机制可能是与结肠黏膜促炎因子TNF-α、IL-6的下调有关。

Abstract:Objective : To observe the effect of Terminalia ehebula Retz.decoetion on rats with ulcerative colitis and to study its imnmne mechanism. Methods : Thirty-two Wistar rats were randomly divided into four groups, i.e, the normal control group, the model group, SASP control group and Terminalia ehebula Retz. decoction group, with 8 in each ( 4 males and 4 females ). Acetic acid was used for the establishment of ulcerative colitis model for rats except those in the normal control group. The rats in the medicinal groups were given the eorrespomting drugs by gavage for 7 days while those in the normal control and model groups were treated with normal salt respectively. The levels of TNF-α and IL-6 expressions in the colon tissue were measured by immunohistochemical method. Results : In colon tissue of rats, the expression of TNF-α the Terminalia ehebula Retz. decoction group dropped significantly compared with those in the model group ( P〈0.01 ) . As opposed to the model group, the Terminalia ehebula Retz. decoction group had an obvious decrease of IL-6 ( P〈O.05 ). Conclusions : Terminalia chebula Retz. has certain therapeutic effect on ulcerative colitis, the mechanism of which may be that it inhibits the colonic inflammation through lowering the TNF-α and IL-6 expressions.
