寒性中药栀子对大鼠甲状腺功能及结构的影响

摘    要:目的:研究长时间摄入寒性中药栀子对正常大鼠甲状腺功能和结构的影响。方法:选用清洁级健康Wistar大鼠30只,雌雄各半。随机分为正常组10只,每日给予生理盐水灌胃;PTU组10只,每次给予等量PTu灌胃;栀子组10只,给予栀子水煎液。连续给药41d后处死,腹主静脉取血,分离血清,用ELISA试剂盒检测三碘甲腺原氨酸(T3)、甲状腺素(T4)、促甲状腺激素(TSH)、促甲状腺激素释放激素(TRH)、甲状腺过氧化物酶(TPO);甲状腺取材,固定、包埋、HE染色,镜下观察。结果:与空白对照组相比,栀子组与PTU组T3、TtKH、TPO均显著降低(P〈0.05),差异具有统计学意义。结论:长时间摄入寒性中药栀子能明显降低正常大鼠T3、TRH、TPO水平,并导致甲状腺的微细结构损伤。

Abstract:Objective: To analyze and discuss the effects of prolonged intake of gardenia on the function and microstructure of thyroid gland in rats. Methods : 30 rats composed by half male and half female were randomly divided into control group, Gardenia group, and PTU group, with 10 rats in each group. Intragastric administration of Gardenia was carried out in accordance with 10 mL/kg body weight. Meanwhile, the control group and PTU group were given intragastric administration of normal salt and PTU in accordance with 10 mL/kg body weight, respectively. The operating of gastric garage was carried out for 40 days. The levels of T3, T4, TSH, TRH and TPO were detected with ELISA method. The HE staining was introduced to observe the microscopic changes. Resuhs : The levels of T3, TRH and TPO decreased dramatically compared with those of the control group ( P〈0.05 ). The pathological changes were observed in the Gardenia group and PTU group. Conclusion : Prolonged intake of Gardenia could lead to hypothyroidism and destroy the structure of thyroid gland.
