半夏与附子不同配伍比例对大鼠毒性研究

摘    要:目的:研究附子与半夏不同比例配伍对大鼠心、肝、肾毒性及形态学的影响。方法:健康SD大鼠60只随机分为空白组、附子与半夏1:1组、附子与半夏1:2组、附子与半夏2:1组、附子组、半夏组,每组1(1只。每组分别按2.5g/(k2·d)灌胃给予生理盐水、附子与半夏1:1煎剂、附子与半夏1:2煎剂、附子与半夏2:1煎剂、附子煎液、半夏煎液。第11天停药后称重,取心脏、肝脏、肾脏标本,计算体重增长率,心、肝、肾脏器系数并观察其形态学变化。结果:各组体重增长率,心、肝、肾脏器系数及心、肾形态学各组间无明显差异,仅1:1组、1:2组、2:1组肝病理切片较其余3纽比较肝细胞稍水肿。结论:附子与半夏配伍煎剂较附子、半夏单煎剂相比心、肾毒性无明显增加,可能具有肝毒性,且1:1、1:2、2:1三种比例间肝脏毒性无显著差异。

Abstract:Objective: To investigate the effects of different compatibility proportions of Aconite and Pinellia on the function and morphology of heart, liver and kidney in rat model. Methods: Sixty SD rats were randomly divided into six groups (n=10), including control group, Aconite and Pinellia 1 : 1 group, Aconite and Pinelli a 1 : 2 group, Aconite and Pinellia 2 : 1 group, the aconite group, and Pinellia group. Each group was given drugs as the dose of 2.5 g/kg per day for 10 days. Mice of the six groups were measured weight and sacrificed on the 11 th day when no drugs were given, and the heart, liver and kidney of mice were taken out and measured weight to calculate the organ coefficient. The morphological changes were observed too. Results: There was no significant difference in weight increase, heart, liver and kidney coefficients, and heart and kidney morphology among groups. In comparison to the control group, in the aconite group and the Pinellia group, a slight hepatocyte edema was observed in the 1 : 1, 1 : 2,2 : 1 groups. Conclusion: Compared with the Aconite decoction and Pinellia decoction, compatibility decoction of Aconite and Pinellia has no significant increase in renal and heart toxicity, but it may have liver toxicity, and no significant difference has been observed among the proportions of 1 : 1,1 : 2,2 : 1 in the aspect of liver toxicity.
