不同含量绞股蓝皂苷对高脂血症大鼠血脂的影响

摘    要:目的:探讨不同含量绞股蓝皂苷对高脂血症模型大鼠血脂水平的影响。方法:采用60只正常雄性SD大鼠,随机分为正常对照组、高脂血症模型对照组、辛伐他汀组及不同含量绞股蓝皂苷(含量分别为40%、80%、98%)组,每组10只;高脂乳剂灌胃8周造模,造模同时分别进行相应药物干预8周。每2周检测一次大鼠血脂及丙氨酸氨基转移酶(ALT)、天门冬氨酸氨基转移酶(AST)的水平。结果:实验大鼠2周时高脂血症模型已复制成功;造模给药4周后,与模型对照组比较,不同含量绞股蓝皂苷(40%、80%、98%)组胆固醇(TC)、低密度脂蛋白(LDL-C)均显著降低(P0.01,P0.05);造模给药6周后,与模型对照组比较,不同含量绞股蓝皂苷(40%、80%、98%)组TC、LDL-C、ALT均显著降低(P0.01);造模给药8周后,与模型对照组比较,不同含量绞股蓝皂苷(40%、80%、98%)组TC、LDL-C、ALT均显著降低(P0.01,P0.05)。不同含量绞股蓝皂苷(40%、80%、98%)组的大鼠血清TC及LDL-C的含量与其相应的组别分别进行相关性分析,造模给药4周、6周后,相关系数r1与r2值在0.3~0.4之间(P1、P20.05);造模给药8周后,相关系数r1与r2值在0.4~0.6之间(P1、P20.01)。结论:不同含量绞股蓝皂苷(40%、80%、98%)均能有效降低高脂血症大鼠血脂水平并与皂苷的含量呈负相关,对肝组织有一定的保护作用,且绞股蓝皂苷98%含量组改善更明显。

Abstract:Objective: To investigate the effect of different contents of gypenoside saponins on blood lipid in rats with hyperlipidemia. Methods: Sixty male SD rats were randomly divided into control group, model group, Simvastatin group and different contents of gynostemma saponins (40%, 80%, 98%) groups, with 10 rats in each group. The models were established by high fat emulsion intragastrically, the models were intervened with corresponding medicine for 8 weeks. We detected the levels of blood lipid, ALT and AST every 2 weeks. Results: After 2 weeks of continuous oral administration of high fat emulsion, the hyperlipidemia models were successful established. After 4 weeks medication, compared with model control group, the TC and LDL-C in gynostemma saponins groups were reduced significantly ( P 〈 0.01, P 〈 0.05 ) ; after 6 weeks medication, compared with model control group, the TC, LDL-C and ALT in gynostemma saponins groups were reduced significantly (P 〈 0.01 ) ; after 8 weeks of medication, compared with model control group, the TC, LDL-C and ALT in gynostemma saponins groups were reduced significantly ( P 〈 0.01, P 〈 0.05 ). We analyzed the contents of TC and LDL-C in gynostemma saponins groups, after 4 weeks and 6 weeks medication, the values of rI and r2 between 0.3 and 0.4 ( P~, P2 〈 0.05 ) ; after 8 weeks of medication, the values of r~ and r2 between 0.4 and 0.6 ( P~, P2 〈 0.01 ). Conclusion : Different contents of gypenoside saponins can reduce the blood lipid level of hyperlipidemia and has negative correlation with the contents of saponin. Gypenoside saponins have positive effect on liver tissue, especially 98% gypenoside saponins.
