| 摘 要： | 目的:探讨同一温度不同时间和同一时间不同温度加热对凋亡坏死、survivin、caspase基因表达的影响与作用机制.方法:对数生长期胃癌AGS细胞,分别在同一温度不同时间和同一时间不同温度加热,用流式细胞仪检测细胞凋亡百分比和坏死百分比,用RT-PCR检测survivin和caspase基因的表达.结果:加热后细胞凋亡增加,加热温度42℃和加热时间2h最为明显.随加热温度升高和加热时间增加坏死增加.加热明显加强Casepase-3的表达和降低Survivin的表达,且42℃加热不同时间时,2h效果最明显,不同温度加热1h时42℃效果最明显.结论:加热能明显抑制胃癌细胞生长,增强Casepase-3的表达和降低Survivin的表达,促进胃癌细胞的凋亡;加热温度42℃、加热时间2h为胃癌细胞热疗的最佳条件.
