摘    要:目的建立同时测定利胆排石片中和厚朴酚、大黄素、厚朴酚、大黄酚和大黄素甲醚5种有效成分含量的HPLC方法。方法采用Diamonsil C18色谱柱(4.6 mm×200 mm,5μm)进行分离,流动相为乙腈-体积分数0.05%磷酸溶液(体积比62∶38),流速为1.0 mL.min-1,检测波长为254 nm,柱温30℃,进样量20μL。结果和厚朴酚、大黄素、厚朴酚、大黄酚和大黄素甲醚的质量浓度分别在0.76~7.6 mg.L-1(r=0.999 2,n=6)、9.4~94 mg.L-1(r=0.999 3,n=6)、0.50~5.0 mg.L-1(r=0.999 5,n=6)、21.0~210 mg.L-1(r=0.999 0,n=6)和5.5~55 mg.L-1(r=0.999 1,n=6)内与峰面积呈良好的线性关系;平均加样回收率依次为98.2%、99.3%、98.0%、98.4%和98.9%,RSD依次为1.9%、1.6%、2.0%、1.9%和1.7%。结论该方法快速、简便、重复性好,适合于同时测定利胆排石片中和厚朴酚、大黄素、厚朴酚、大黄酚和大黄素甲醚的含量。

Abstract:Objective To develop an RP-HPLC method for the determination of honokiol,emodin,magnolol,chrysophanol and physcion in Cholagogic and lithagogue(traditional Chinese medicines)tablets.Methods A Diamonsil C18 column(4.6 mm×200 mm,5 μm)was used at 30 ℃.A acetonitrile-0.05% phosphoric acid aqueous was used as the mobile phase with isocratic elution(V∶ V=62∶ 38).The flow rate was set at 1.0 mL · min-1.The detection wavelength was set at 254 nm.Results The linear response ranged from 0.76-7.6 mg · L-1 for honokiol(r=0.999 2,n=6),9.4-94 mg · L-1 for emodin(r=0.999 3,n=6),0.50-5.0 mg · L-1 for magnolol(r=0.999 5,n=6),21.0-210 mg · L-1 for chrysophanol(r=0.999 0,n=6)and 5.5-55 mg · L-1 for physcion(r=0.999 1,n=6),respectively.Recoveries were 98.2% with RSD 1.9% for honokiol,99.3% with RSD 1.6% for emodin,98.0% with RSD 2.0% for magnolol,98.4% with RSD 1.9% for chrysophanol and 98.9% with RSD 1.7% for physcion.Conclusions The method is quick,simple and repeatable for simultaneous determination of honokiol,emodin,magnolol,chrysophanol and physcion in Cholagogic and lithagogue tablets.
