5-氟脲嘧啶腹腔化疗对鼠胃肿瘤细胞凋亡的实验研究

摘    要:目的通过对种植性胃肿瘤鼠腹腔注射5-氟脲嘧啶(5-Fu)化疗,探讨5-Fu腹腔化疗对鼠胃肿瘤细胞凋亡的影响。方法利用Walker-256细胞株建立鼠种植性胃肿瘤模型,将40只Wister雄性胃肿瘤大鼠随机分为2组,生理盐水(对照组)和5-Fu组。接种后七天,对照组每只每天腹腔注射生理盐水40ml/k9,5-Fu组每只每天腹腔注入5-Fu20mg/k9,两组均连用5天,于化疗结束后第七天每组处死10只。观察:肿瘤体积(肿瘤体积抑制率),Tdt介导的DNA末端原位标记染色法(TUNEL法)检测细胞凋亡,IMS细胞图象分析系统自动测量凋亡细胞面积及光密度,余下10只观察生存时间,并计算生存时间延长率。结果生理盐水组动物于接种后2周出现食量明显减少,活动少,倦怠状,对外界刺激反应迟钝,逐渐消瘦,甚至腹水形成,而5-Fu组接种后4周才出现,与生理盐水组相比,5-Fu组鼠胃肿瘤生长明显缓慢,肿瘤体积明显减少(P〈0.05),肿瘤抑制率为36.57%(P〈0.05),生存时问明显延长,延长率为23、7%(P〈0.05)。肿瘤细胞凋亡面积增大,光密度明显升高。结论5-Fu有明显的诱导胃肿瘤细胞分化与凋亡作用。

Abstract:Objective To study effect on apoptosis to Implanted Gastric Tumor in Rats with 5-fluorouracil(5-Fu) by Intraperitoneal Chemotherapy(IPC).Methods 40 Wistar rats were implanted gastric tumor with the malignant tumor cell line Walker-256 and then divided randomly into 2 groups: 0.9% sodium chloride(NS) group and 5-Fu group. At the end of the 7th day after IPC, 10 rats in each group were executed to test tumor volume. The apoptosis were measured by terminal deoxynuleotidy1 transferase mediated nick and labeling (TUNEL) methods, and its area and by optical density processed automatically with the IMS cell image analyze system. The rest of the rats were observed untill their death to calculate the survive time extension ratio.Results The tumor volume in the 5-Fu group dwindled in comparison with NS group(P<0.05). The survival time in the 5-Fu group was obviously longer than that in NS group(P<0.05). The apoptosis area and optical density in the 5-Fu group was significantly increased compared with NS group(P<0.05).Conclusion 5-FU can induce apoptosis and inhibit the tumor growth.
