摘    要:目的 研究姜黄素对化疗药物环磷酰胺所致微核形成的影响,测定了小鼠血清超氧化物歧化酶、丙二醛、总抗氧化能力、还原型谷胱甘肽等指标的变化。方法 50只小鼠随机分成5组,即正常组,模型组,高剂量组(50mg/kg),中剂量(10mg/kg),低剂量(2mg/kg)。正常组即阴性对照组,只喂基础饲料;模型组为环磷酰胺阳性对照组,单次尾静脉注射环磷酰胺1mL/只,浓度为100mg/kg;试验组在环磷酰胺给药前后24h尾静脉注射姜黄素各1次1mL/只,浓度分别为50、10、2mg/kg。末次给药24h后将小鼠拉颈处死,每只小鼠取股骨1枚作微核观测,并眼眶取血,经离心分离制得小鼠血清作其他指标检测。结果 姜黄素对环磷酰胺所引起的微核发生率有着显著的抑制作用。血清中超氧化物歧化酶活性和丙二醛含量明显降低,总抗氧化能力和还原型谷胱甘肽含量明显提高。结论 提示姜黄素不仅能改善环磷酰胺在肿瘤治疗中所导致的氧化还原状态的失衡,而且能减少由环磷酰胺所诱发的微核形成。

Abstract:Objective To investigate the effects of curcumin on micronuclei formation frequency induced by cyclophosphamide. Method The content of malondialdehyde(MDA) was measured by thiobarbituric acid(TBA) method,the activities of superoxide dismutase(SOD),the levels of reduced glutathione(GSH),and the total antioxidantive capacity(T-AOC) were detected by spectrophotography.Mice were divided into five groups,the control group was treated with saline,model group was treated with cyclophosphamide alone,and the treated group was injected by cyclophosphamide combined with curcumin.1 mL of cyclophosphamide at 100 mg/mL was given only once to each mouse.The dosage of curcumin was 100 mg/kg,10 mg/kg,and 2 mg/kg body weight,respectively.Results The results showed that curcumin was able to reduce the micronuclei formation frequency induced by cyclophosphamide,and the activity of T-AOC and the content of GSH increased,while SOD and MDA decreased in plasma.Conclusion These results obtained from our experiments suggest that curcumin had protective effects against micronuclei formation frequency induced by cyclophosphamide.Further studies on the pharmaceutical functions and immunological responses of curcumin may help the development of its clinical application.
