猪组织工程皮肤自体及异体移植后血管重建的实验研究

摘    要:目的研究贵州小型香猪来源的组织工程皮肤自体及同种异体移植后血管形成情况,为复方壳多糖组织工程皮肤临床应用提供实验依据。方法按我科已建立的方法构建猪的组织工程皮肤,然后进行自体及同种异体移植实验,观察移植的猪组织工程皮肤的存活及创面愈合情况。并于移植后1、4、8周取标本,行组织学检查和免疫组化染色,观察自体及同种异体移植后猪组织工程皮肤血管形成状况。结果猪组织工程皮肤自体及同种异体移植后均存活,创面达到理想的愈合效果,组织学检查显示新生血管丰富;免疫组化染色显示:Ⅷ因子免疫组化染色阳性。结论本实验构建的猪组织工程皮肤自体及同种异体移植后,可获得理想的创面愈合效果,猪组织工程皮肤血管形成良好,与正常皮肤相似。本实验为复方壳多糖组织工程皮肤临床应用提供了有力的实验依据。

Experimental study on reconstruction of the blood vessles after autograft and allograft of swine composite chitosan tissue engineered skin
Abstract:Objective To provide experimental evidence for clinical application of composite chitosan tissue engineered skin,a swine tissue engineered skin was fabricated to study reconstruction of the blood vessles after autograft and allograft.Methods Serially passaged cells were used to fabricate composite chitosan tissue engineered skin which had been reported by authors. Then the composite chitosan tissue engineered skin were autografted and allografted onto full thickness excised wounds of two swines after organotypic culture in vitro.It's viability and wounds healing status were observed.Biopsy of the grafts sampled at week 1st,4th and 8th respectively were examined by histologically and immunohistochemically so as to investigate reconstruction of blood vessles of tissue engineered skin.Results The tissue engineered skin were living after autograft and allograft,merged with the surrounding normal skin of the swine.The wounds healed well.The tissue engineered skin transplanted onto the swine showed immunohistochemically that factor Ⅷ was expressed in dermis.Conclusion Wounds heal well after autograft and allograft of the tissue engineered skinthe angiogenesis of tissue engineered skin is well, as like normal skin.This animal study provides experimental evidence for clinical application of composite chitosan tissue engineered skin.
