丹参多酚酸纯化工艺研究

摘    要:目的:优选丹参多酚酸的纯化工艺条件。方法:采用高效液相色谱法,以丹酚酸B的含量为指标,对絮凝剂和大孔吸附树脂纯化条件进行了考察。结果:最佳工艺条件丹参多酚酸提取液按每克药材加入约1%壳聚糖醋酸溶液1ml,絮凝2h,萃取pH为2~3,采用D101大孔吸附树脂,上样量药材:树脂为0.5g:1g,以5BV50%乙醇溶液洗脱,丹参酸B含量达85.50%。结论:该工艺可较好的纯化富集丹参多酚酸有效成分。

Abstract:Objective : To study the preparative purification of salvianolie acid from salvia mihiorrhiza. Method: The optimum preparative purification of macroreticular resin and flocculant was evaluated by measuring the content of Salvianolic acid B by HPLC. Restilt: The optimum conditions were screened,which were lml 1% chitosan corresponding to 1g salvia mihiorrhiza, and flocculated for 2h, and the volume of 50% (V/V) ethanol as eluant was 5BV. By this method, the elution efficiency of salvianolic acid B exceeded 90%. Conclusion: The method is more effective for large-scale isolation and purification of salvianolie acid from salvia mihiorrhiza.
