肿瘤细胞多药耐药的发生机制

摘    要:多药耐药(MDR)是指肿瘤细胞对一种抗肿瘤药物产生耐药的同时,对化学结构和作用机制完全不同的其他抗肿瘤药物产生交叉、广谱耐药的现象。MDR是导致肿瘤化疗失败的主要原因之一,目前已涉及到临床常用的多种抗肿瘤药物,其发生机制复杂,由多分子、多途径介导。本文就药物转运蛋白、酶类、5-氟尿嘧啶代谢酶学异常等因素介导肿瘤细胞发生MDR的机制作一综述。

Abstract:Multidrug resistance denotes that the tumor cells have cross resistance to various anticancer drugs which differ in chemical structure and mode of action. It is the main cause of failure of cancer chemotherapy. Multidrug resistance has been involved in many commonly used anticaneer drugs, its mechanism is complicated and is mediated by multiple molecular pathways. This article reviewed the mechanism of multidrug resistance in tumor ceils mediated by drug transporters, enzymes, and metabolic enzyme abnormalities of 5-fluorouracil, etc.
