DNA多态性分析凝胶干胶保存法及其法医学应用价值

目的：探讨凝胶干胶保存法及其在法医学中的应用价值。方法：采用饱和酚／氯仿抽提法提取血祥DNA，STR—PCR进行扩增，6％变性聚丙烯酰胺凝胶垂直电泳，聚丙烯酰胺凝胶经银染显色后，采用玻璃纸处理制成干胶。结果：将银染后的聚丙烯酰胺凝胶及时进行拍照同时将其制成干胶，用Marker做对照通过测量DNA带的位置发现所得干胶同原始胶及拍照获取胶的结果一致。结论：通过玻璃纸干胶保存法可使原始资料永久而真实的保存，此方法对法医物证的保存具有极其重要的意义。

The method to preserve dry gel of DNA polymorphism and its application in forensic medicine

Objective:To study the method of preserving dry gel and its application in forensic medical.Methods:Genomic DNA was abstracted using phenol-chloroform from fresh blood samples.DNA samples was amplified by short tendem repeat-PCR.PCR products were separated on 6% denaturing polyacrylamide gel.After silver stain the polyacrylamide gel was preserved by glass-paper.Results:We found genotypes from dry gel conserved by glass-paper were same as original gel preserved by photograph after polyacrylamide gel were taken picture and made dry gels in time.Simutanously the position of belt of DNA were tested with DNA marker.Conclusion:Original data of forensic medical can be preserved by dry gel forever and truthfully.Therefore the method of dry gel plays important role in forensic medicine.
