新型微囊系统体外培养原代肝细胞的研究

摘    要:目的 探讨一种新型微囊系统以体外培养原代肝细胞。方法 使用新型微囊系统体外培养大鼠原代肝细胞,检测微囊系统各项特性,检测肝细胞的生存率、尿素合成和白蛋白合成功能。结果 做囊具有良好的通透能力,白蛋白可自由通过,具有一定的机械稳定性,并具有良好的免疫保护能力,分子量在150kD以上的物质不能进入做囊,肝细胞在微囊内保持较高的活性和功能,其尿素合成和白蛋白合成功能比对照组高1~3倍。结论 此微囊具有良好的牛物和机械特性,可以在生物性人工肝和细胞移植中发挥重大作用。

Abstract:Objective To investigate a new microcapsular system for culture of primary generation hepatocytes in vitro. Methods A novel microcapsular system was used for in vitro culture of primary generation rat hepatocytes .The various characteristics of the microcapsular system were examined. The survival rate of hepatocytes and the functions of urea and albumin synthesis were observed.Results The microcapsule had good permeability,a certain degree of mechanical stability and favorable immunological protective ability. Hepatocytes within the microcapsule maintained a high degree of viability and function, and the synthesis of urea and albumin was 1~3 times higher than that of controls.Conclusions This microcapsule has excellent biological and mechanical properties.This system can be of great benefit for use in bioartificial liver and cell transplantation.
