变应性鼻炎大鼠鼻黏膜Bcl-2蛋白表达与细胞凋亡的研究

摘    要:目的探讨变应性鼻炎(AR)大鼠鼻黏膜Bcl-2蛋白的表达与细胞凋亡之间的关系,进一步了解变应性鼻炎的发病机制。方法选健康SD大鼠20只,雌雄不限,随机分为实验组和对照组,各10只。实验组以卵清蛋白腹腔注射致敏,继之鼻局部激发建立变应性鼻炎大鼠模型。取实验组与对照组动物鼻呼吸区黏膜,行Bcl-2蛋白免疫组织化学染色,以细胞凋亡末端脱氧核苷酸转移酶介导的dUTP缺口末端标记(TUNEL)法检测鼻黏膜细胞凋亡状况,以图像分析软件分析结果。结果变应性鼻炎大鼠鼻黏膜Bcl-2蛋白主要表达于细胞胞质中,包括鼻黏膜腺体细胞、上皮细胞,固有层中的淋巴细胞,鼻分泌物中可见大量呈强阳性反应的嗜酸性粒细胞,实验组Bcl-2蛋白表达水平显著高于对照组。凋亡细胞散在于鼻黏膜上皮层和固有层,实验组更多见于上皮层,实验组凋亡指数与对照组相比下调。结论变应性鼻炎大鼠鼻黏膜细胞凋亡过程受抑可能是变应性鼻炎鼻分泌物增多及上皮破坏的机制之一。

Abstract:Objective To study the effect of Bcl-2 protein in apoptosis on nasal mucusa of rat in allergic rhinitis(AR)model. Methods Twenty SD rats weighing 180-220 g, free of nasal diseases, were randomly divided into two groups. AR model was established through intraperitoneal injection of ovalbumin(OVA) as well as nasal challenge with OVA. The nasal mucusa thus obtained from two groups were studied by HE, immunohistochemical staining of bcl-2 protein and terminal deoxynucleotidyl transferase-mediated dUTP nick endlabeling (TUNEL). Results The positive expression of Bcl-2 was presented in the glandular cells, eosinophil, Lymphocytes and epithelium cells . The staining density of Bcl-2 protein in AR rats increased significantly as compared with those in normal controls. Apoptosis cell scattered in nasal mucusa. Apoptotic index down-regnlated in AR rat. Conclusion The suppression of apoptosis maybe play an important role on the gland hyperplasia and injury of epithelium of AR rats.
