SMAF1基因的异位过表达对脂肪细胞3T3-L1增殖和凋亡的影响

摘    要:目的观察一脂肪细胞特异性表达的新基因小脂肪细胞因子1(SMAF1)在小鼠脂肪前体细胞3T3-L1中的异位表达对脂肪细胞增殖和凋亡的影响。方法构建小鼠SMAF1真核表达载体并稳定转染3T3-L1脂肪前体细胞以获得永久异位表达SMAF1基因的细胞株,使用Westernblot证实SMAFl基因的表达。体外培养SMAF13T3-L1脂肪细胞,以MTT法检测SMAF13T3-L1脂肪细胞的增殖;用流式细胞仪对SMAFl3T3-L1脂肪细胞进行凋亡分析。结果与对照相比,SMAF13T3-L1脂肪细胞增殖明显降低,差异有统计学意义(P〈0.05);而SMAF13T3-L1脂肪细胞凋亡分析结果与对照无显著性差异。结论SMAFl基因在3T3-L1中的异位性表达,对3T3L1前体脂肪细胞生长增殖有显著的抑制作用,而对细胞凋亡无影响,提示SMAF1在脂肪细胞增殖分化中发挥一定的作用。

Effect of ectopic expression of SMAF1 on proliferation and apoptosis of preadipocyte 3T3-L1

Abstract:Objective To evaluate the effect of a novel adipocyte-specific expressed gene small adipocyte factor 1 (SMAF1) on proliferation and apoptosis of preadipcyte 3T3-L1. Methods A permanent SMAF1 expression construct was established for stable transfection of 3T3-L1 and formation of SMAF1 3T3-L1 cell line. After invitro cul turing of cells, both MTT method and flow cytometry were used to detect and analyze the proliferation level and apoptosis progress respectively. Results Compared with control groups, SMAF1 3T3-IA cell line showed remarkable reduction in proliferation with statistical significance (P〈0.05), while no notable difference in apoptosis between two cell lines. Conclusion The ectopic expression of SMAF1 in 3T3-L1 preadipocyte might suppress proliferation without interfering apoptosis process.
