大鼠创伤失血性休克模型的成功建立与论证

摘    要:目的探讨建立理想的创伤失血性休克模型的方法。方法 40只健康SD大鼠随机分为对照组和休克组。休克组在完成股动脉、股静脉插管后用电刀在腹正中切开约5 cm长的切口,通过动脉放血,在10 min内将血压维持在休克水平[平均动脉血压(MAP)降到40 mm Hg],约1 h后进行液体复苏0.5 h,而对照组在同等条件下不进行创伤和失血实验,而是观察1.5 h。实验结束后两组大鼠均拔出留置针,结扎血管并缝合切口,给予室温25℃保温、饲养。72 h后处死大鼠,检测大鼠肺、肠组织的水含量,用RT-PCR法检测肺、肠组织细胞间黏附分子-1(ICAM-1)的mRNA表达量,并做病理切片。数据采用SPSS 10.0软件处理,两组间死亡率的比较采用χ2检验,而两组间的肺、肠组织水含量及ICAM-1 mRNA表达量比较采用t检验,P〈0.05为差异有统计学意义。结果休克组与对照组比较,肺、肠组织水含量[肺:(0.808±0.020)vs(0.557±0.024);肠:(0.748±0.028)vs(0.503±0.026)]、肺、肠组织ICAM-1 mRNA与-βactin的电泳灰度比值差异均有统计学意义(P均〈0.05)。结论大鼠创伤失血性休克模型出现了类似临床上创伤失血性休克后的肺、肠组织损伤的表现,并有一定的死亡率。

Abstract:Objective To establish the rat model of traumatic and ensanguined shock.Methods Forty healthy SD rats were randomly assigned to the control group and the shock group.The abdomens of rats in the shock group were cut 5 cm length with radio knives and let bleeding to MAP(around 40 mm Hg)within 10 minutes,which remained for 1 hour.The rats were then rescued with fluid for half an hour.Rats in control group were cut but no further trauma or bleeding.All rats were killed after maintaining body temperature at 25℃ for 72 hrs.The water-containing level of rats′ lung and ileum tissue was evaluated with drying box.The mRNA of ICAM-1 was tested by RT-PCR for all the rats.Results There were significant differences of water-containing levels in lung(0.808±0.020 vs 0.557±0.024)and ileum tissue(0.748±0.028 vs 0.503±0.026)between the rats in the shock and the control groups(P0.05).The expression levels of ICAM-1 were also different between the two groups(P0.05).Conclusion The rat model of traumatic and ensanguined shock was established successfully.
