双荧光探针实时RT-PCR检测不同样品中甲型H1N1病毒核酸的临床意义

摘    要:目的采用双荧光探针实时RT-PCR方法检测甲型H1N1流感病毒感染患者不同样品中H1N1病毒核酸,了解其对甲型H1N1流感的快速诊断及病程进展的关系。方法用双荧光探针实时RT-PCR法分别检测118例H1N1患者发病48 h后和(或)恢复期咽拭子、血浆及下呼吸道分泌物中H1N1病毒核酸。结果 118例H1N1患者发病48 h后咽拭子H1N1病毒核酸阳性;57.5%(23/40)重症患者下呼吸道分泌物中H1N1病毒核酸阳性,恢复期3例阳性;37.5%(15/40)重症患者血浆中检测到H1N1病毒核酸。结论双荧光探针实时RT-PCR法可以快速检测H1N1病毒感染,血浆中H1N1病毒核酸阳性可能是重症患者的标志。

Abstract:Objective To investigate the positive proportion of H1N1 influenza A RNA testing in different clinical specimens.Methods Real-time RT-PCR was used to test H1N influenza A viral RNA in the throat swabs,blood plasma and lower respiratory tract secretions of 118 patients with dual fluorescent probes.The specimens were collected at 48 hrs of onset of the disease as well as at the period of convalescence.Results One hundred and eighteen throat swab specimens were positive for H1N1 influenza A RNA testing after 48 hrs of onset of the disease.57.5%(23/40)of lower respiratory tract secretions and 37.5%(15/40)of plasma of severe patients were positive for H1N1 influenza RNA,respectively.Three plasma were found positive for patients at the period of convalescence.Conclusion The real-time RT-PCR with dual-fluorescent probes can quickly detect H1N1 RNA.The positive results in blood plasma might be an indicator of severe cases.
