加味桃核承气汤及其不同提取物对糖尿病大鼠心肌细胞钙转运的影响

摘    要:目的研究加味桃核承气汤及其不同提取物对糖尿病大鼠心肌细胞钙转运作用的影响。方法将加味桃核承气汤原方通过水提醇沉、正丁醇、乙酸乙脂不同处理方法制备不同提取物。清洁级SD大鼠用STZ注射复制糖尿病模型,随机分为加味桃核承气汤全方、水提醇沉、正丁醇、乙酸乙脂和达美康、模型、空白7个组,分别用上述药物灌胃治疗8周,采用定磷法检测心肌细胞Na -K -ATP酶和Ca2 -ATP酶活性。结果除全方组外糖尿病各组Na -K -ATP酶活性和Ca2 -ATP酶活性下降,其中模型组下降最为明显,其次是达美康组,而全方组明显上升,与模型组、达美康组相比差异均非常显著(P<0.01),其余中药各组虽有下降但不甚明显。上述各组中,阻止Na -K -ATP酶活性下降的效果依次为:全方组>正丁醇组>乙酸乙脂组>水提醇沉组>达美康组;而阻止Ca2 -ATP酶活性下降的疗效依次为:全方组>乙酸乙脂组>正丁醇组>水提醇沉组>达美康组。结论加味桃核承气汤全方及其正丁醇、水提醇沉、乙酸乙脂等不同提取成分均有提高或稳定糖尿病大鼠心肌细胞Na -K -ATP酶和Ca2 -ATP酶活性的作用,因而在一定程度上可以阻止糖尿病大鼠的心肌肥大和心功能下降的病理改变。除全方组外,其提取物中以正丁醇和乙酸乙脂组的疗效为优。

Effect of Modified Taohe Chengqi Decoction and its Extracts on Calcium Transport of Cadiocyte in Diabetic Rats

Abstract:Objective To investigate the effects of supplemented Taohe Chengqi Decoction and its different extraction components on calcium transport of cadiocyte in diabetic rats. Methods The extracts of Modified Taohe Chengqi Decoction were prepared and extracted by water-alcohol, n-butyl alcohol, and ethyl-acetate. Diabetic rat model was induced in clean (CL) Sprague Dawley rats (SD rats) by single intraperitoneal injection of 55 mg/kg body weight of streptozotocin, and these were randomly divided into 7 groups:the decoction group, extracted groups by water-alcohol, n-butyl alcohol, and ethyl-acetate, Diamicron treatment group, model rats group and normal group. Drugs above-mentioned were lavaged in for 8 weeks, and detected Na -K -ATPase activity and Ca2 -ATPase activity of cardiac muscle cell by the colorimetric phosphorus determination method. Results The activity of Na -K -ATPase and Ca2 -ATPase decreased in model rats group, Diamicron treatment group, water-alcohol, n-butyl alcohol, and ethyl-acetate group, of which model group was the most significant, and followed the diamicron group. Inversely, that was obviously increased in the whole decoction group, which showed significant difference compared with model group or with Diamicron group (P <0.01). The sequence of effect on inhibiting Na -K -ATPase activity was as followed:whole-decoction group>n-butyl alcohol group>ethyl-acetate group>water-alcohol>Diamicron group. The sequence of effect on inhibiting Ca2 -ATPase activity was as followed:whole-decoction group>ethyl-acetate group>n-butyl alcohol group>water-alcohol>Diamicron group. Conclusion Modified Taohe Chengqi Decoction and its diferent extractions maybe effective on raising or stabilizing the activity of Na -K -ATPase and Ca2 -ATPase in cadiocyte of diabetic rats, so it could prevent myocardium hypertrophy and patho-change by heart function failure in variably extent. Ethyl-acetate group and n-butyl alcohol group showed rather good effect among these extractions.
