泄浊解毒方对慢性非特异性溃疡性结肠炎大鼠内皮细胞黏附分子表达的影响

摘    要:目的探讨泄浊解毒方对慢性非特异性溃疡性结肠炎(CUC)大鼠内皮细胞黏附分子(ICAM-1)表达的影响。方法将40只SD大鼠随机分为5组,每组8只,即空白对照组、模型对照组、阳性药对照组和泄浊解毒方高、低剂量组。空白对照组正常喂食水,其余4组采用三硝基苯磺酸(TNBS)/乙醇溶液灌肠法制作CUC大鼠模型。空白对照组予0.9%氯化钠注射液灌胃,其余4组均于造模后第3d灌胃给药。模型对照组予0.9%氯化钠注射液灌胃,阳性药对照组予柳氮磺胺吡啶0.30g/kg灌胃,泄浊解毒方高、低剂量组分别予泄浊解毒方相应剂量(4、1mL/d)灌胃,均每日1次,共14d。给药结束后观察各组大鼠一般状态、光镜下大鼠结肠黏膜组织形态和各组疾病活动指数(DAI)、ICAM-1表达的变化。结果阳性药对照组和泄浊解毒方高、低剂量组DAI、ICAM-1积分均较模型对照组下降(P0.01);泄浊解毒方高剂量组DAI较阳性药对照组、泄浊解毒方低剂量组均下降(P0.01,P0.05)。阳性药对照组和泄浊解毒方高、低剂量组ICAM-1积分均较模型对照组降低(P0.01);泄浊解毒方高、低剂量组ICAM-1积分均较阳性药对照组降低(P0.05);泄浊解毒方高、低剂量组之间ICAM-1积分比较差异无统计学意义(P0.05)。结论泄浊解毒方对TNBS诱导CUC大鼠疗效确切,泄浊解毒方高剂量组优于阳性药对照组,其作用机制可能与通过下调ICAM-1表达有关。

Effect of Xiezhuo Jiedu decoction on expression of ICAM-1 of rat with chronic nonspecific ulcerative colitis

Abstract:Objective To study the effect of Xiezhuo Jiedu decoction on expression of ICAM-1 of rat with chronic nonspecific ulcerative colitis induced by trinitrobenzene sulfonic acid(TNBS).Methods Ulcerative colitis rats were induced by intragastric administration of trinitrobenzene sulfonic acid(TNBS).40 SD rats were randomly divided into 4 groups:model group,Xiezhuo Jiedu Decoction high dose group,Xiezhuo Jiedu Decoction low dose group,positive control group,and 10 rats were chosen to be blank control group before model making.The rats in all the groups were administered by intragastric administration.The rats in model group and blank control group were administered by 0.9% sodium chloride injection,the rats in positive control group were given Sulfasalazine(0.30 g/kg),the rats in Xiezhuo Jiedu Decoction high dose group and Xiezhuo Jiedu Decoction low dose group were given Xiezhuo Jiedu Decoction stock solution(4 mL/d,1 mL/d),all once a day.Intragastric administration was begun 3days after model-making and lasted 14 days.18 days after model-making,the changes of morphology and histology of colonic mucosa,disease active index(DAI) and the rats'ICAM-1 were observed in all the groups.Result The scores of DAI and ICAM-1 in positive control group,Xiezhuo Jiedu Decoction high dose group,and Xiezhuo Jiedu Decoction low dose group were decreased in comparison with those in control group(P〈0.01).The change of DAI in Xiezhuo Jiedu Decoction high dose group was superior to that in Xiezhuo Jiedu Decoction low dose group and positive control group(P〈0.05,P〈0.01).The score of ICAM-1 Xiezhuo Jiedu Decoction high dose group and Xiezhuo JieduDecoction low dose group was lower than that in positive control group(P〈0.05).There was no significant difference between Xiezhuo Jiedu Decoction high dose group and Xiezhuo Jiedu Decoction low dose group on the score of ICAM-1(P〈0.05).Conclusion Xiezhuo Jiedu Decoction has definite effect on ulcerative colitis rats induced by trinitrobenzene sulfonic acid(TNBS),and Xiezhuo Jiedu Decoction high dose group is superior to positive control group.The mechanism of action was related to down-regulate the expression of ICAM-1.
