摘    要:目的:研究腮腺多形性腺瘤和瘤旁组织中差异表达的基因。方法:分别从5例腮腺多形性腺瘤组织和与其相对应的瘤旁组织中提取总RNA并纯化为mRNA,两种不同组织的mRNA分别逆转录合成有荧光分子标记的探针,然后与Agilent人全基因组芯片进行杂交,杂交信号用Agilent扫描仪扫描,结果用ImgGene 3.0软件分析。结果:腮腺多形性腺瘤和瘤旁组织有差异表达的基因447个,其中表达上调185个,下调262个。结论:运用基因表达谱芯片可以筛选出腮腺多形性腺瘤相关基因。

Abstract:Objective: The purpose of this study was to screen the differentially expressed genes between salivary pleomorphic adenomas and their surrounding normal tissues. Methods: The total RNAs were isolated from five pleomorphic adenomas and their adjacent normal tissues, and their mRNAs were purified. Both the mRNAs from two kinds of tissues were reversely transcribed to cDNAs with the incorporation of fluorescent-labelled dUTP to prepare the hybridization probes. The mixed probes were hybridized to the whole human genome of Agilent genechips. Tumor related genes were screened through the analysis of fluorescent intensity. Results: In five cases with pleomorphic adenoma and the tissue adjacent pleomorphic adenoma, 447 genes were screened out for differentiation in their expressed level, among which 185 were upregulated and the other 262 downregulated. Conclusion: cDNA microarray technique may be effective to screen the related genes in pleomorphie adenoma. Futher analysis of those differentially expressed genes may be useful to explore the molecular mechanism of pleomorphic adenoma.
