| 摘 要： | 目的:建立一种能同时测定龙胆泻肝片中栀子苷和黄芩苷含量的高效液相色谱法。方法:采用Ultimate C18色谱柱,流动相为甲醇(A)-0.2%磷酸溶液(B),梯度洗脱为0~20min,22%A;20~21min,22%~44%A;21~40min,44%A,检测波长为254nm,流速1.0mL/min,柱温为30℃。结果:该色谱条件下栀子苷峰和黄芩苷峰之间有良好的分离度,栀子苷在3.096~61.92μg/ml范围内线性关系良好,r=0.9998,平均回收率为98.43%,RSD为1.22%(n=6)。黄芩苷在8.52~213μg/ml范围内线性关系良好,r=0.9999,平均回收率为97.97%,RSD为1.35%(n=6)。结论:本方法快速简便,准确可靠,灵敏度高,可作为提高龙胆泻肝片质量标准的参考。

| Abstract: | Objective ： A HPLC method was established for determination of two Glycosides composition in Longdan Xiegan Tablets. Methods ： A Ultimate CI8 column was used with methanol （A）and 0.2% phosphoric acid （B）as the mobile phase. Gradient elution：0-20min,22%A;20- 21min, 22%-44%A;21-40min, 44%A. The flow rate was 1.0mL/min at 30～C and the detector wavelength was 254nm. Result： The relationshiop between the concentration and the peak area of gentiopicrin,geniposide,baicalin was good linear relation.The calibration curve of geniposide was at the range of 3.096 to 61.92μg/ml （r=0.9998）,and the recovery was 98.43 and RSD=1.22%（n=6）. The calibration curve of baicalin was at the range of 8.52 to 213μg/ml （r=0.9999）,and the recovery was 97.97 and RSD =1.35%（n=6）. Conclusion：The method was rapid,sensitive and ac- curate,and could be used for a reference to enhance quality control of Longdan Xiegan tablets. |
