激活粒细胞对大鼠离体心脏再灌注损伤的实验研究

摘    要:本实验采用大鼠离体心脏模型,探讨激活粒细胞对大鼠离体心脏再灌注损伤的作用机制。实验过程观察冠状动脉流量,冠状动脉阻力,冠状动脉流出液中乳酸脱氢酶的活性及再二醛的含量。实验完毕后对心肌进行组织学检查及超微结构观察。

Abstract:solated rat hearts were reperfused with phorbol myristate acetate (PMA)activated polymorphonu-clear leucocytes(PMNs)(n =10),polymorphonuclear leucocytes(PMNs)(n=10)and Krebs-Hensleit(K-H)(n=10)to observe coronary flow,coronary resistence lactate dehydrogenase(LDH),malondi-adehyde(MDA) of coronary fluid and morphologic changes by light and electronic microscope. The re-sults showed PMA-activated PMNs decreased coronary flow,increased coronary resistence,LDH andMDA. Morphologic changes also showed that PMA-activated PMNs caused severe injury. It is conclud-ed that activated PMNs cause reperfusion injury mediated by oxygen free radicals released from activat-ed-PMNs. The obstruction of microcirculation by activated PMNs increases reperfusion injury.
