内皮祖细胞与颈动脉粥样硬化斑块性质的关系

摘    要:目的探讨外周血内皮祖细胞数量和功能的变化与颈动脉粥样硬化斑块性质的关系。方法选择确诊有颈动脉粥样硬化的脑梗死患者以及无颈动脉粥样硬化的健康体检者。所有入组人员均行颈动脉彩色多普勒超声检查以及头颅计算机X射线断层扫描或磁共振成像检查。根据颈动脉超声病理改变结果分为3组:软斑块组、硬斑块组和对照组。从肘静脉采血10 mL,用密度梯度离心法获取单个核细胞,将其接种在人纤维连接蛋白包被的24孔培养板,培养4天后,洗去非贴壁细胞,换液培养至7天。收集贴壁细胞,采用激光共聚焦显微镜鉴定,FITC标记荆豆凝集素I和Dil标记的乙酰化低密度脂蛋白双染阳性细胞为正在分化的内皮祖细胞。采用黏附能力测定实验、MTT法和改良的Boyden小室分别观察内皮祖细胞的黏附能力、增殖能力和迁移能力。结果放大200倍视野下内皮祖细胞数量、黏附能力,硬斑块组和软斑块组均高于对照组,软斑块组高于硬斑块组,差异均具有统计学意义。硬斑块组和软斑块组增殖能力均低于对照组,软斑块组高于硬斑块组,差异具有统计学意义。软斑块组迁移能力高于对照组,差异具有统计学意义。结论外周血内皮祖细胞的数量和功能可能与颈动脉粥样硬化斑块性质有关。
