弥漫大B细胞淋巴瘤中B7-H1临床表达水平分析

目的 对弥漫大B细胞淋巴瘤(DLBCL)中B7-H1的表达水平进行分析.方法 收集DLBCL患者病理活检或手术切除的肿瘤组织标本50例,另收集瘤周正常淋巴组织标本50例作为对照.采用Western Blot法检测B7-H1在瘤周正常淋巴组织与DLBCL肿瘤组织中的表达水平;采用流式细胞仪检测瘤周正常淋巴组织与DLBCL肿瘤组织中B7-H1和调节性T细胞(Treg)的表达水平.结果 Western Blot检测结果显示,B7-H1在DLBCL肿瘤组织与瘤周正常淋巴组织中均有表达,与瘤周正常淋巴组织比较,DLBCL肿瘤组织中B7-H1的表达水平较高.HLA-DR+CD14+巨噬细胞在DLBCL肿瘤组织中表达B7-H1的比例较瘤周正常淋巴组织高(P﹤0.05).与瘤周正常淋巴组织相比较,DLBCL肿瘤组织中(包括IPI值≥2分与IPI值﹤2分)pDC、Treg、mDC与调节性T细胞(CD4+/CD8+)的比例较高(P﹤0.05).结论 B7-H1在DLBCL患者的淋巴组织中呈高表达,提示B7-H1与淋巴瘤细胞的免疫逃逸有关,从而造成了DLBCL细胞免疫平衡的失调.

Analysis of the clinical expression level of B7-H1 in diffuse large B-cell lymphoma

Objective To analyze the clinical expression level of B7-H1 in diffuse large B-cell lymphoma (DLBCL). Method 50 cases of tumor tissue samples were collected by pathologic biopsy or surgical resection in DLBCL patients, and 50 cases of normal lymphatic tissue samples were obtained as control. Western blot was used to detect the expression of B7-H1 in normal lymphatic tissues and DLBCL tumor tissues. Flow cytometry was applied to determine the expres-sion of B7-H1 and regulatory T cells (Treg) in normal lymphatic tissues and DLBCL tumor tissues. Result The results of Western Blot showed that B7-H1 expression was observed in DLBCL tumor tissues and normal adjacent lymphatic tis-sues, and the expression level of B7-H1 in DLBCL tumor tissues was higher than that in normal adjacent lymphatic tis-sues. The proportion of HLA-DR+CD14+macrophages expressing B7-H1 in DLBCL tumor tissues was higher than that in normal adjacent lymphatic tissues (P<0.05). Compared to the normal adjacent lymphatic tissues, the proportion of pDC (including that with IPI≥2 and IPI<2), Treg, mDC, and regulatory T cell (CD4+/CD8+) was also higher in DLBCL tumor tissues (P<0.05). Conclusion B7-H1 protein was highly expressed in the lymphatic tissues of patients with diffuse large B-cell lymphoma, suggesting that B7-H1 was associated with immune escape of lymphatic cells, resulting in the imbal-ance of immunologic regulation of DLBCL cells.
