实验性骨质疏松大鼠骨组织内Ⅰ型胶原mRNA的表达水平

摘    要:目的 为了探讨在骨质疏松发生过程中骨合成的改变,检测去势大鼠骨组织Ⅰ型胶原mRNA的表达水平。方法 切除SD大鼠双侧卵巢,复制骨质疏松动物模型,从骨组织内提取RNA,以地高辛标记的点杂交方法检测去势和对照组Ⅰ型胶原mRBA的表达。结果 去势大鼠骨组织Ⅰ型胶原mRNA的滴度较对照组明显降低。结论骨质疏松发生过程中有风Ⅰ型胶原mRNA表达的下降,提示骨质疏松不仅涉及骨吸收,还有骨合成的降低。

Abstract:Objective To examine the change of bone formation on osteoporosis, the expression level of type I collagen mRNA in normal and ovariectomized rats bone Methods Female SD rats were ovariectomized as models of experimental osteoporosis, RNA samples were extracted in bone tissues, the expression level of type I collagen mRNA were detected using digoxigenin dUTP labeled dot blotting hybridization Results The expression levels in normal group were significantly higher than those in ovariectomized rats Conclusion The expression level of type I collagen mRNA in osteoporosis bone tissue was lower It is suggested that the formation phase of bone metabolism, as well as the resorption phase, play an important role in the pathogenesis of osteoporosis
